Objective:To investigate the distribution of Methylenetetrahydrofolate reductase(MTHFR)genotypes of subjects in Chongqing city of China,and explore the relationship between MTHFR C677T polymorphisms and carotid artery plaque(CAP)?Intracranial atherosclerotic stenosis(ICAS).Methods:Six hundred ninety eight ethnic Han Chinese in Chongqing were selected per sex ratio stratified sampling.Medical histories were obtained and ultrasound examinations were performed on the arteries.Polymerase chain reaction-restriction fragment length polymorphism and Sanger sequencing genotyping assays was used to detect MTHFR C677T rs 1801133 genotypes.Results:A significant difference in distribution between the CAP and NCAP group of allele frequencies(C/T +T/T and C/C,? 2=14.434,P<0.001)?ICAS and ICAS group of allele frequencies(C/T +T/T and C/C,?2=13.347,P<0.001)were detected.There was a significant difference at the genotype(T/T and C/C)and(C/T and C/C)groups,The frequency of T allele in the ethnic population with CAP and ICAS was higher than those of control as expected.Binary stepwise logistic regression analysis demonstrated that T/T genotype and C/T genotype were independent risk factors for carotid artery plaque and Intracranial atherosclerotic stenosis.Conclusion:The C677T genetic polymorphism of the MTHFR gene in the Han population of Chongqing is associated with an increased risk of intracranial and extracranial atherosclerosis.
