Objective and background:Recent researches had revealed that heat shock protein plays an important role in tumor development and metastasis. The research was to show and analyze the expression and significance of heat shock protein105 in pancreatic carcinoma.Methods:Expression of HSP105 mRNA in human pancreatic carcinoma was detected by reverse transcription-polymerase chain reaction, using Western-blot method to detect the protein expression level of HSP105 in human pancreatic cancer, HSP105 express by immunohistochemistry in cell lines and human pancreatic carcinoma tissue , expression of HSP105 was present by immunohistochemistry in human pancreatic carcinoma tissue and matched normal tissue.Result:HSP105 mRNA and protein express in two types of pancreatic carcinoma cell lines (Panc-1 and Bxpc-3).The positive rate of HSP105 in pancreatic carcinoma was 87.5%(35/40), while the rate of the protein in matched normal tissue was 12.5%(5/40).The difference was significant (p<0.05). HSP105 was correlated with tumors differentiation and metastasis (p<0.05).Conclusions: HSP105 may play an important role in the differentiation and metastasis of pancreatic carcinoma. There was a relation between the protein expression and the malignancy degree of pancreatic carcinoma, which will be considered valuable in evaluating the ontogenesis and prognosis of the human pancreatic carcinoma. And HSP105 will possibly become a new therapeutic target.
