Objective:To investigate the expression of hMLH1 and hMSH2 protein in hypopharyngeal carcinoma and normal laryngeal tissue,then reveal their relationships and prognosis between hMLH1 and hMSH2 protein with hypopharyngeal carcinoma. So as to provide the theoretical basis for prevention,early diagnosis,judge prognosis and genetic treatment of hypopharyngeal carcinoma.Methods :In this study, 50 cases were randomly selected from 2008 to 2010,and those were selected by different stages of surgical treatment of hypopharyngeal cancer. Rrom the surface of the adjacent 3 mm, 5 mm, 10 mm,each tissue were made into paraffin sections . Then took 20 cases of normal laryngeal tissues compared with them, by using immunohistochemical SP method, the expression of hMLH1 and hMSH2 protein in hypopharyngeal carcinoma,margin and normal laryngeal tissue were deteced.From this were can explore their relationships and prognosis between hMLH1 and hMSH2 protein with hypopharyngeal carcinoma.Results:1.Positive expression of hMLH1 and hMSH2 protein in hypopharyngeal carcinoma and normal laryngeal tissue was mainly localized in nucleus, and the color was brown or yellow,but fews were localized in cytoplasm ,and the color was superficial uniform.2. The positive expression of hMLH1 in hypopharyngeal carcinoma and normal laryngeal tissue were 54% and 80%,and there was significant difference between them(P<0.05).The positive expression of hMSH2 in hypopharyngeal carcino- ma and normal laryngeal tissue were56% and85%,and there was significant difference between them (P<0.05).3.The expression of hMLH1and hMSH2 in hypopharyngeal carcinoma had no relationship with gender,age and clinical stage of tumor (P>0.05). 4.The expression of hMLH1and hMSH2 in hypopharyngeal carcinoma were related with differentiation level and metastasis of lymphonode (P<0.05).5. There was a positive correlation bettween the expression of hMLH1 and hMSH2 in hypopharyngeal carcinoma,r=0.713(P<0.05).6.The positive expression of hMLH1and hMSH2 had increased in sequence of cancer tissue of those at 3mm,5mm,10mm adiacent carcinoma and normal mucosa.The nearer to the normal tissue,the more the proteins were expressed (P<0.05).Conclusions:1.The protein of hMLH1 and hMSH2 gene were down regulated in hypopharyngeal carcinoma cell,they may participate the early occurrence of hypopha- ryngeal carcinoma.2.The expression of hMLH1 and hMSH2 in hypopharyngeal carcinoma had no association with gender,age and clinical stage of tumor.3.The expression of hMLH1 and hMSH2 in hypopharyngeal carcinoma was related with differentiation level and metastasis of lymphonode.4.The was a positive correlation bettween the expression of hMLH1 and hMSH2 in hypopharyngeal carcinoma, hMLH1 and hMSH2 may play an important role in the occurrence,development, invasion and metastasis of hypopharyngeal carcin- oma.5.hMLH1 and hMSH2 protein may be a tumor marker in the occurrence of hypopharyngeal carcinoma.6.Examining the expression of gene hMLH1 and hMSH2 protein in hypopharyngeal carcinoma,they may provide theoretical basis for the occurrence,deve- lopment and genetic treatment of hypopharyngeal carcinoma.7.The excision margin at 10mm away from the tumor can be regared as a safe range for surgery of hypopharyngeal carcinoma.
