Expression At Protein Level And Clinical Significance Of Mismatch Repair Gene HMLH1 And HMSH2 In Nasopharyngeal Carcinoma

Objective: By detecting the expression of the mismatch repair (MMR) gene hMSH2 and hMLH1 in 50 cases of nasopharyngeal carcinoma and 30 case of normal mucosa in pharynx nasalis, investigate their effect on the mechanism of nasopharyngeal carcinoma (NPC) on protein level and the relationship between the two genes and the clinical stage of NPC, expecting to provide thinking and methods for the early stage diagnostics and the therapy.Methods: Nasopharyngeal carcinoma tissues were obtained from 50 cases nasopharyngeal carcinoma patients, normal mucosa in pharynx nasalis from 30 cases normal tissue of nasopharynx as controls. The expression of hMSH2 and hMLH1 were detected by Elivision TMS-P immunohistochemistry.Result: The mismatch repair (MMR) genne hMSH2 and hMLH1 highly express in 30 cases of normal mucosa in nasopharynx. In 50 cases of nasopharyngeal carcinoma, hMLH1 positive rate is 84.0% and hMSH2 positive rate is 22.0%.Compared to normal mucosa in nasopharynx, the discrepancy of hMSH2 is obvious and has the significant statistical significance(P<0.01). hMLH1 has no statistical significance discrepancy(P > 0.05). There is no dependability between the two genes and clinical stage(P>0.05). Besides the sex, the level of expression above 50 years old and below 50 years old has no statistical significance(P> 0.05).Conclusion: The abnormal expression of hMSH2 in normal mucosa of nasopharynx and NPC has obvious discrepancy, hinting that it might be involved in the invasion of NPC. Maybe hMLH1 has no connection with the invasion of NPC. There is no relation between the expression of the two genes and the age, and the same to the sex. And maybe the immunohistochemistry could quickly and efficiently help doctor for early diagnosis and treatment.
