Background:With the increase of older people, the incidence of cardiac-cerebral thrombosis disease is increasing, and cardiac-cerebral thrombosis disease has become one of the most common causes of death. Thrombosis is the pathological process of multiple factors and steps, including the mechanism of vascular factors, blood coagulation, anticoagulation and fibrinolysis disorders. Revealing the pathogenesis of thrombosis, the clinical diagnosis and treatment of thrombosis has an important significance. Growth arrest-specific gene 6(Gas 6) is a vitamin K dependent protein family members, co-ligand of Axl family. Recently a small amount of literature at home and abroad has been reported that the plasma Gas 6 levels and gene polymorphism is related to cardiac-cerebral thrombosis disease, but results vary. This is the first in the Chinese population to explore the correlation of plasma Gas 6 level and gene polymorphism with cardiac-cerebral thrombosis disease and its possible mechanism, for providing a theoretical basis of prediction, diagnosis and treatment of linical cardiac-cerebral thrombosis disease.Objectives:1. To reveal the correlations of plasma Gas 6 levels and cardiac-cerebral thrombosis disease (cerebral infarction, acute myocardial infarction).2. To reveal the relationships of Gas 6 gene polymorphism and cardiac-cerebral thrombosis disease (cerebral infarction, acute myocardial infarction). 3. To explore the possible relationship and mechanism between plasma Gas 6 levels and its intron c.834 +7 G> A gene polymorphism.4. To fill the blank of study on Gas 6 with cardiac-cerebral thrombosis disease gene polymorphism in Chinese population.Methods:1. The blood samples in patients with cerebral infarction, cerebral hemorrhage, acute myocardial infarction and healthy control group were collected.2. The plasma Gas6 levels and high sensitivity C-reactive protein (hs-CRP) levels in patients with cerebral infarction, cerebral hemorrhage, acute myocardial infarction and healthy control group were determined by Enzyme-Linked ImmunoSorbent Assay.3. The genomic DNA was extracted from white cells in each group of blood samples.4. To amplify the genomic DNA by Polymerase Chain Reaction (PCR), and identify Gas 6 gene polymorphism by gene sequencing.Results:1. The plasma Gas 6 level was significantly higher (P <0.01) in patients with cerebral infarction, cerebral hemorrhage and acute myocardial infarction than that in healthy controls.2. The Gas 6 level in AA genotype of the Gas 6 in intron c.834 +7 G> A was significantly decreased (P <0.01) compared with normal controls, but GA genotype was higher (P <0.01).3. The AA genotype and A allele frequency of Gas 6 c.834 +7 G> A was significantly lower (P <0.01) in patients with cerebral infarction, cerebral hemorrhage, acute myocardial infarction than in healthy controls.4. The plasma hs-CRP levels were significantly higher (P <0.01) in patients with cerebral infarction, cerebral hemorrhage and acute myocardial infarction than that in controls, and Gas 6 levels were synchronized with hs-CRP levels.5. The cerebral infarction, cerebral hemorrhage and acute myocardial infarction were significantly(P <0.01)related to these risk factors of smoking, hypertension, hyperlipidemia and diabetes.Conclusions:1. The plasma Gas 6 levels were correlated with cerebral infarction, cerebral hemorrhage, and acute myocardial infarction.2. The plasma Gas 6 levels of the AA genotype among Gas 6 gene c.834 +7 G> A locus AA, GG and GA genotypes were negative correlation with cerebral infarction, cerebral hemorrhage, and acute myocardial infarction, indicating that the level Gas 6 of the AA genotypes may reduce the risk of thrombotic disease by way of downregulating expression in thrombotic diseases.3. The AA genotype and A allele frequency of Gas 6 gene c.834 +7 G> A was negatively correlated with cerebral infarction, cerebral hemorrhage and acute myocardial infarction, reflecting the AA genotype as a protective risk factors for cerebral infarction, cerebral hemorrhage and acute myocardial infarction.4. Gas 6 may be simila with the hs-CRP, as an acute phase protein, and increase in acute phase response of cerebral infarction, cerebral hemorrhage and acute myocardial infarction.5. The cerebral infarction, cerebral hemorrhage and acute myocardial infarction were significantly related to these risk factors of smoking, hypertension, hyperlipidemia and diabetes.
