Association Of Human PARL Gene Rs3732581, Rs73887537 Polymorphisms With Type 2 Diabetes Mellitus In Gansu Han And Hui Population

Objective To investigate association of the rs3732581,rs73887537 polymorphisms of the PARL gene with type 2 diabetes mellitus (T2DM) and T2DM-related quantitative traits in Gansu Han and Hui nationality respectively. To investigate distribution difference of genotype and allele frequencies of the two polymorphisms of PARL between Han and Hui nationality.Methods We genotyped PARL gene rs3732581,rs73887537 polymorphisms in 181 patients with T2DM and 128 healthy controls of Han nationality and in 136 patients with T2DM and 98 healthy controls of Hui nationality by using PCR-RFLP technique.Anthropometric parameters were recorded.Plasma glucose, insulin, lipid, creatinine and urea nitrogen levels were measured by biochemical technique. Homa-IR,BMI and WHR were calculated. Based on the previous reported studies our analysis was to compare GG genotype versus CG/CC genotype.Statistical analysis was performed using SPSS,Version 11.0.Results 1.We did not find rs73887537 polymorphism of PARL gene was existed in both studied Gansu Han and Hui nationality.2. There was no significant difference in PARL rs3732581 polymorphism distribution between Han and Hui nationality(P >0.05).3.PARL rs3732581 polymorphism genotype frequencies of GG,CG, CC were 13.3%,48.1%,38.7% in type 2 DM group and 18.8%,46.9%,34.4% in control group, allele frequencies of G,C were37.3%,62.7% in type 2 DM group and 42.2%, 57.8% in control group in Han nationality respectively. There was no significant difference in frequencies of genotype and allele between the two groups in Han nationality (both P>0.05).4.PARL rs3732581 polymorphism genotype frequencies of GG, CG, CC were 11.8%,49.3%,39.0% in type 2 DM group and 17.3%,48.0%, 34.7% in control group, allele frequencies of G, C were 36.4%,63.6% in type 2 DM group and 41.3%,58.7% in control group in Hui nationality respectively. There were no significant differences in frequencies of genotype and allele between the two groups in Hui nationality (both P>0.05).5. The subjects carrying GG genotype had higher plasma triglyceride level than that of the subjects carrying CG and CG+CC genotype. The subjects carrying GG genotype had higher diastolic blood pressure than that of the subjects carrying CG and CC and CG+CC genotype. Urea nitrogen level in subjects carrying GG genotype was lower than that of the subjects carrying CG and CG+CC genotype (P<0.05) in T2DM group of Han nationality.6. The subjects carrying GG genotype had higher plasma triglyceride level than that of the subjects carrying CG+CC genotype.The subjects carrying GG genotype had higher diastolic blood pressure than that of the subjects carrying CG and CC and CG+CC genotype. High-density lipoprotein cholesterol level in subjects carrying GG genotype was lower than that of the subjects carrying CG+CC genotype (P<0.05).Urea nitrogen level in subjects carrying GG genotype was lower than that of the subjects carrying CG and CG+CC genotype (P<0.05) in T2DM group of Hui nationality.7. The subjects carrying GG genotype had higher plasma triglyceride level than that of the subjects carrying CC and CG+CC genotype (P<0.05) and total cholesterol level in subjects carrying GG genotype was higher than that of the subjects carrying CG and CC and CG+CC genotype (P<0.05) in control group of Han nationality.8. There was no significant difference on clinical phenotype between different genotypes of PARL rs3732581 polymorphism in control group of Hui nationality.Conclusion 1.We did not find rs73887537 polymorphism of PARL gene was existed in both studied Gansu Han and Hui nationality.2. There was no significant difference in PARL rs3732581 polymorphism distribution between Han and Hui nationality.3. The rs3732581 polymorphism of PARL gene is not associated with T2DM in both Gansu Han and Hui nationality. However, it is associated with lipid metabolism and diastolic blood pressure and urea nitrogen level in T2DM group of Gansu Han and Hui nationality and associated with lipid metabolism in control group of Gansu Han nationality.
