Objective:To investigate the expression and clinical significance of the hTERC gene in cervical lesions. Methods:The expression of hTERC gene in tissues from 20 normal cervical exfoliated cells,100 cervical exfoliate cell with intraepithelial neoplasia in cytologic diagnosis and 10 cases of histological specimen diagnosed cervical squamous cell carcinoma,were detected by the fluorescent in situ hybridization (FISH). Results:Positive expression rates of the hTERC gene in normal cervixes,ASC,LSIL and HSIL were 0.00%,30.00%,41.86% and 71.43%. Compared with normal cervixes and LSIL,HSIL, hTERC gene positive expressions in LSIL,HSIL were significantly different. The positive expression rates of the hTERC gene in normal cervixes,CIN1,CIN2-3 and cervical squamous cell carcinoma were12.50%,60.00%,69.23%,100.00%. Compared with normal cervixes and CIN, hTERC gene positive expressions in CIN were significantly different, and there was a significant difference in cervical squamous cell carcinoma and control cervix. Conclusion:Our study suggested that the over expression of hTERC gene may play an important role in cervical carcinogenesis, therefore it can be used as a better tumor index for early cervical cancer screening diagnosis. Object:Detecting the expression of p16,Ki67,Cytokeratin8,Cytokeratinl7 in cervical intraepithelial neoplasia (CIN) and invasive squamous cell carcinoma tissue, and exploring the pathological significance. Methods:By using immunohistochemical method, we examined the expression of p16,Ki67,CK8,CK17 in 44 cases of cervical intraepithelial neoplasia (20 cases of CIN1,24 cases of CIN2-3),25 cases of invasive squamous cell carcinoma and 20 cases of normal cervical tissue. Results:The expression of p16,Ki67,CK8,CK17 were all negative in normal cervical epitheliums. Positive expression rates of p16,Ki67,CK8,CK17 in invasive cervical squamous cell cancer were 100%,100%,88%,92%, p16 is strongly expressed, Ki67,CK8,CK17 is medium and strongly positive; in CIN-H were 100%,100%,58.3%,58.3%, p16 is medium and strongly positive, Ki67,CK8,CK17 is medium positive; in CIN-Lwere 65%,65%,10%,30%, all with weak and medium positive. Conclusion:The positive expressions of p16,Ki67,CK8,CK17 were closely related to consecutive cervical lesions. p16,Ki67,CK8 and CK17 play an important role in the progress of carcinomatous change.
