Objective:to study the expression of minichromosome maintenance proteins7and nuclear antigen KI67protein in the normal cervical tissue and other different pathological lesion levels of cervical tissue, discuss their clinical significance and mechanism in the evolution of cervical lesions, and analyzes their relationship of the expression in cervical squamous cell carcinoma.Methods:the72cases of specimens were come from the cervical biopsy and surgical removal of the fresh tissue samples during2012-2013annual, all the specimens have been confirmed by pathology, and they did have no chemical, immunization, radiation or other therapy treatment. According to the international general diagnosis standard they were be divided into four groups, including:cervical normal group, CIN I group, CINⅡ-Ⅲ group and cervical squamous carcinoma group; According to the international union of gynecology and obstetrics of2009(FIGO) staging surgery cervical squamous carcinoma group can be divided into:phase I a-b and the phase II a-b; According to the histologic,the cervical squamous cell carcinoma can be divided into three groups:high differentiated squamous carcinoma group, differentiated squamous carcinoma group, low differentiated squamous carcinoma group; According to the average age (24-61years old) cervical squamous carcinoma group can be divided into:40years of age group or less,>40years old group. Using immunohistochemical SABC method and real-time PCR method respectively to detect the expression of MCM7and KI67rate (amount) in cervical normal group, CIN I group, CIN Ⅱ-Ⅲ group and cervical squamous carcinoma,then statistical analysis to investigate their clinical significance and relationship between them in the evolution of the cervical cancer.Results:using the different methods to detect the expression in cervical tissue, the results are basically identical.1.The results of immunohistochemical method:1)MCM7protein and KI67protein positive expression in cervical lesions were located in the nucleus, and show to be claybank. MCM7protein and KI67protein in cervical normal group, the CIN I group, CIN Ⅱ-Ⅲ group and cervical squamous cell carcinoma, the positive expression rate showed an increasing trend, respectively:29.4%,60%,83.3%,93.9%;23.5%,50%,50%,91%, the difference was statistically significant in the four groups (P<0.05).2) In the cervical squamous cell carcinoma, the positive expression rate of of MCM7protein in Ⅱa-b phase is higher than that of I a-b phase, the difference was statistically significant in the two groups (P<0.05), related to the pathological classification, comparison among the three groups the difference was statistically significant (P<0.05),unrelated to the age, the difference was no statistically significant difference in the two groups (P>0.05);The expression of KI67protein in cervical squamous cell carcinoma is related to its pathologic stage, the difference was statistically significant in the three groups (P<0.05), the expression of KI67protein is not related to age and clinical stage, the difference was statistically unsignificant in the two groups(P>0.05),there was no statistically significant difference with ages (P>0.05).2. Rt-PCR results:1) The expression mass of MCM7mRNA and KI67mRNA in the four groups of were gradually increasing, respectively is:1.00±0.00,2.24±0.78,3.07±1.01,4.49±1.3,41.00±0.00,1.03±0.12,1.18±0.08,1.48±0.24, the difference was statistically significant in the four groups (P<0.05).2) In the cervical squamous cell carcinoma, the expression of MCM7mRNA is associated with clinical staging and pathologic stage, the difference was statistically significant in the two groups (P<0.05), unrelated to the age, the difference was not statistically significant in the two groups (P>0.05); the expression of KI67mRNA is associated with pathologic stage, the difference was statistically significant in the three groups (P<0.05), but unrelated to age and clinical stage, there was no statistically significant difference (P>0.05).3. In cervical squamous cell carcinoma, the expression rate (amount) of MCM7protein (mRNA) and KI67protein (mRNA) protein were positively correlated (P<0.05).Conclusion:The expression rate (amount) of MCM7protein (mRNA) is increasing with the severity of disease in cervical tissue, and the expression is associated with clinical stages and pathological classification, and has nothing to do with age in cervical squamous carcinoma, that prompt MCM7participate in cervical squamous cell carcinomas of the evolution process,being one of the important factors to promote the cervical squamous carcinoma.KI67protein (mRNA) in cervical tissues is also increasing with the severity of disease,the expression in cervical squamous carcinoma is closely related to the pathological grading and clinical staging,but independent of the age, that prompt KI67closely associated with the incidence of cervical squamous carcinoma.MCM7and KI67expression in cervical squamous cell carcinomas were positive correlation relationship, they jointly promote the occurrence of cervical squamous carcinoma.Because the important function and significance of MCM7and KI67in cervical cancer and precancerous lesions, so the detection their expression in cervical lesions, it provide reliable diagnostic basis for the cervical cancer occurrence, evolution and so on.
