Objective: Cervical cancer is one of the primary malignant tumor that serious threat to women's health. It's incidence rate can be the second malignant tumors of female reproductive system . Every year there is about 0.493 million new cases and 0.274 million deaths. In developing countries, cervical cancer can be the first one among the multiple gynecological tumor . China's newly discovered cases per year for 0.1315 million. In the past few decades,the Chinese women's cervical cancer screening carried out and lead cervical cancer mortality decreased, but every year there are still a lot of new cases. Therefore ,reduceing cervical cancer morbidity and mortality, screening and monitoring of prognosis of precancerous lesions is the key.In this study, we use fluorescence in sit- uhybridization to detect the expression of telomerase in varying degrees cervical scraping samples and to evaluate the effect and clinical significance of hTERC in cervical lesion.Methods: For the first clinical hospital of Jilin University from March 2008 to March 2009,we examined 115 cases patients who acceptance of screening patients admitted to hospital for treatment or before treatment. Into the standard: In accordance with cervical intraepi- thelial neoplasia diagnosis treatment norms, sub-standard based on pathological diagnosis of CINⅠ,Ⅱ,Ⅲ-class is divided into cervical cancer diagnosis in accordance with the "Obstetrics and Gynecology," the sixth edition of the diagnostic criteria. Using fluorescence in situ hybridization detected 20 cases of normal, 63 cases of cervical intraepithelial neoplasia and 32cases of uterine cervix cancer cells in the expression of hTERC.Results:Positive expression rates of the hTERC gene are 15%, 61.9%, 63. 64% and 93. 75%.There is significant difference between CINⅠand CINⅡ-CINⅢ(P<0.01). Compared with normal control and CINⅠ, positive expression of the hTERC in cervical cancer are significantly difference(P<0.01). Cervical cancer group and CINⅡ-Ⅲgroup hTERC gene expression rates, there are significant difference (P<0.05); hTERC gene expression in CINⅡ-Ⅲgroup and normal control group there is significant difference (P<0.01). The percentage of comparative analysis of hTERC gene amplification of each group show that: CINⅠgroup 8.00±2.00%; CINⅡgroup 11.00±2.00%; CINⅢgroup 15.00±10.00%; cancer group 28.50±29.29%. The comparation between Cancer group and CINⅡ-Ⅲgroup have significant difference (P <0.05); The average gene copy number of hTERC show that: CINⅠgroup 2.08±0.12, CINⅡgroup 2.12±0.06, CINⅢgroup 2.18±0.12, cervical cancer group 2.37±0.39. Cancer group and CINⅡ-Ⅲ more significant difference between the cancer group and CINⅡ-Ⅲgroup compared with significant difference (P <0.05)Conclusion: Study results indicate that the abnormal activation of telomerase may occur at various stages of cervical cancer. With the increase in the degree of histological lesions,the amplification rate and degree of hTERC gene got increased.The hTERC gene of cervical scraping samples could be used as an early screening of cervical cancer, early diagnosis and treatment of auxiliary indicators.
