Observation Of Malondialdehyde, Myeloperoxidase And Antioxidative Indicatrixes In Serum Of Patients With Graves Disease

OBJECTIVE:To observe the change of malondialdehyde(MDA),myelopero- xidase(MPO), total antioxidant capacity(T-AOC),superoxide dismutase activity (SOD) and glutathione peroxidase(GSH-PX) in serum of patients with Graves disease,and to evaluate the antioxidant/oxidant balance in patients with Graves disease.METHODS:?Subjects in the study consisted of two patient groups (45 newly diagnosed patients with overt hyperthyroidism due to Graves disease and 45 euthyroid Graves disease patients who had been treated with antithyroid drug more than 2 months) and one control group (45 healthy patients).Fasting serum sample(more than 8 hours)were analyzed for T-AOC, SOD, GSH-PX, MPO and MDA,and all indicatrixes mentioned above were measured by chemical colorimetric method.RESULTS:The patients with newly diagnosed Graves disease had increased level of MDA in comparison with patients in euthyoid Graves disease(P<0.001) and the control(P<0.001),and the levels of MDA were not different between the patients with euthyoid Graves disease and the control group.The MPO activity increased in both group of patients with newly diagnosed Graves disease (P<0.001) and patients with euthyoid Graves disease(P=0.021), meanwhile the level was higher in patients with newly diagnosed Graves disease compared with patients with euthyoid Graves disease(P<0.001). The SOD levels of patients with newly diagnosed Graves disease were significantly higher than that of control group(P<0.001).The levels of T-AOC, and GSH-PX were not significant different between patients with newly diagnosed Graves disease and control group.The levels of SOD and GSH-PX in newly diagnosed Graves disease patients were the same as that in patients with euthyoid Graves disease,? the difference of T-AOC levels was significant between newly diagnosed Graves disease patients and patients with euthyoid Graves disease (P=0.019). There were no significant differences for serum level of T-AOC, SOD and GSH-PX, between newly diagnosed Graves disease and control group.CONCLUSION:The balance between oxidation and antioxidative defense was disrupted in patients with newly diagnosed Graves disease.Oxidative stress increases in patients with newly diagnosed Graves disease. Oxidative stress is related to hyperthyroidism.The level of myeloperoxidase increases in patients with Graves disease.
