Objective:To evaluate the effect of Danzhi compound on hyperthyroidism and to explore the protective mechanism of Danzhi compound on hyperthyroid liver injury.Methods:The GD model is established by immunizing BALB / c female mice with Ad-TSHRa-289.The mice are divided into normal blank group,model group,low,middle and high dose group and methimazole group.During the administration,the general state,body temperature,body weight,diet and drinking water of the mice are observed.After 6 weeks,the mice serum,thyroid tissue and liver tissue samples were collected.The levels of TSH,T3 and T4 in serum are measured by competitive method,the serum TRAb concentration is measured by biotin double antibody sandwich enzyme-linked immunosorbent assay(ELISA),the thyroid tissue is stained with HE,in order to verify the effect of Danzhi compound.The levels of AST,ALT and ALP in serum ate detected by colorimetric method,the activities of antioxidant enzymes SOD,CAT and DNA damage enzymes 8-OHDG in liver tissue are detected by competitive method,to explore the prevention and treatment mechanism of Danzhi compound on hyperthyroid liver injur.Results:1.In improving the body temperature and diet of hyperthyroid mice,there is no significant difference between Danzhi compound and Methimazole group(P> 0.05).In the weight and drinking water of hyperthyroid mice,Danzhicompound is lower than that of Methimazole group,and the difference is statistically significant(P <0.05).2.TRAb,Danzhi compound middle dose group and high dose group compare with no significant difference between Methimazole group,(P> 0.05);Methimazole group of thyroid function TSH?T3?T4 are normal;Danzhi compound low dose group TSH,T3 are not decreased,T4 decreased;Danzhi compound middle dose group,TSH normal,T3,T4 decreased,but do not resume normal;Danzhi compound TSH of high dose group is increased,but no recovery,T3 is decreased,T4 decreased but do not recover;Danzhi compound in each dose group and Methimazole group,there is significant difference,(P<0.05).3.The thyroid cells of each dose group of Danzhi compound are recovered to varying degrees,But compared with Methimazole group,the situation is slightly worse;4.Methimazole group hyperthyroidism liver injury in mice decreased ALT,but do not return to normal,AST,ALP fail to reduce;Danzhi compound middle dose group,high dose group can restore AST,ALP,ALT to normal;low dose group AST,ALT return to normal,ALP no reduction;5.Methimazole group of SOD,CAT decreased,but do not return to normal,8-OHDG fails to take effect;The levels of SOD,CAT and 8-OHDG in the middle and high dose groups of Danzhi compound are normal,and there is no significant difference between the two groups(P> 0.05).The activity of SOD and CAT in the low dose group of Danzhi compound is improved,but the 8-OHDG returns to normal level.6.On the liver pathological morphology of the repair,Danzhi compound stronger than Methimazole group.Conclusion:1.GD model is successfully reproduced by immunization with BALB / c female mice with Ad-TSHRa-289,and liver injury is observed in mice;2.Danzhi compound reduce the high metabolic symptoms,improve thyroid function,regulate immune disorders,repair thyroid tissue;3.Danzhi compound inhibit hyperoxia liver cell DNA oxidative damage,restore liver function,repair liver tissue.
