Shenqi Fuzheng Injection's Effections On IL-6,TNF-α In Mice With Viral Mycarditis

Objective:To study the influence to the expression of IL-6 and TNF-a on Viral Myocarditis(VMC)in Mice when treated with Shenqi Fuzheng injection,which to provide evidence for clinic.Method:120 4-week-old BALB/c male mice were randomly divided into five groups, twenty five per group, including:normal group(A), viral group(B), Shenqi Fuzheng injection(short by Shenqi following) large dose group(C), Shenqi low dose group(D), ribavirin group(E). B-D groups were infected(0.1ml inteaperitoneally)with CVB3 diluted in saline. 8 of each group were killed on day 3,10,30 after inouculation.Blood were obtained by eye-ball excision, and their hearts were removed sterility andobserved:(1), Histopathologic examination of the heart by optical microscope, myocardial histopathologic scores were counted.(2), Serum IL-6 were measured uantitatively by using ELISA method.(3), Immunohistochemicalq methods was used to detect the protein expression of TNF-α.(4), Reverse-transcription polymerse chain reaction(RT-PCR)was used to detect expression of TNF-αmRNA in myocardium.Result:1, Pathological changes of myocardium:There were no pathological changes in myocardium in group A, and the incidence of myocarditis was 100% in other groups. In group B, the pathological damage was reduced to minimum.2, the expression of ELISA: As compared to infection group,the expression of IL-6 reduced in treatment group(P<0.01); 3,The results of Immunohistochemical methods:It showed that TNF-αin viral group was obviously higher than normal group(P<0.01);Compared with B group, each drug treatment group could decrease the expression of TNF-α. Among these, the expression of Shenqi group were significantly decreased(P<0.01)4,The results of RT-PCR:Compared with normal group, TNF-αmRNA of viral group were significant increased(P<0.01).The expression of TNF-αmRNA in Shenqi group,Shenqi low dose group and ribaviin group were significant lowed than the viral group(P<0.01, P<0.05).Conclusion:IL-6,TNF-αshould be important roles in the pathogenesis of viral myocarditis, Shenqi Fuzheng injection can reduce expression,reduce the changes of ultrastructure and pathology,improve the survive rate and protect myocardial tissue.
