Objective : To investigate the effects of Shenqifuzheng injection on the inflammation factor of mice model with viral myocarditis induced by Coxsackievirus B3, and to explore its mechanism which to provide scientific evidence for clinic.Methods:Utilizing the cardiac tendency of Coxsackie virus B3 to make the model of viral myocarditis in mice, by injecting Coxsackie virus B3 to the peritoneal cavity of the mice.120 Balb/c male mice were divided into 5 groups at random:blank group,virus model group, western group,low dose traditional Chinese medicine group and high dose traditional Chinese medicine group, treat the mice with the Shenqifuzheng injection for 30 days. Then observe the change of Pathomorphology in the cardiacmuscle tissue,the rate of death,the weight change and the cardiac weight. pathological changes of cardiac muscle by HE dyeing,And immunohistochemistry method was used to exam the expression of ICAM-1 and MCP-1. semiquantitative analysis of ICAM-1 and MCP-1mRNA in myocardial by RT-PCR methods.Results:1 Survival rate of mice:The blank group was the highest,the second was Shenqi high dose group and the lowest survival rate is the model group.2 The Pathology change in myocardial: There were no morphology change in the myocardial of Control group,Other experimental groups of mice have different degrees of myocardial pathological changes。Pathology damage Shenqifuzheng highl dose group is lighter than Shenqifuzheng low dose group , control group and model group3 The expression of MCP-1 and ICAM-1 protein: the results of immunohistochemical staining showed that the expression of protein ICAM-1 and MCP-1 in model group were all obviously higher than control group(P<0.01), Compared with model group, each drug treatment group can decrease the expression of MCP-1 and ICAM-1 Among these, the expression of Shenqi high dose group was significantly decreased(P<0.01)4 The results of RT-PCR: compared with control group, MCP-1 and ICAM-1mRNA of model group were significant increased(P<0.01)The expression of MCP-1 and ICAM-1mRNA in small dose and medicine groups were lower than the model group(P<0.01, P<0.05);high dose group were significant lower than the model group(P<0.01)Conclusion:1: Shenqifuzheng injection can inhibit the toxic effects of the heart caused by coxsackie virus.2: Shenqifuzheng injection can reduce myocardial necrosis.3: Shenqifuzheng injection can decreased the expression of MCP-1 and ICAM-1 or inhibit their activity.
