Objective:The aim of the study to investigate the expression and significance of mRNA and protein of MCP-1 and CCR2 the alteration of human kidney tissues of patients with Hydronephrosis caused by unilateral ureteral obstruction.Methods:Collect renal tissue samples from patients with varying degrees of hydronephrosis. The dynamic change of messenger RNA expression of monocyte chemotactic protein-1 (MCP-1) and CCR2 were evaluated by using quantitative, real-time, fluorogenic reverse transcription polymerase chain reaction, R-T PCR (TaqMan) monitoring assay. Immuno-fluorescin histochemistry method and Western-Blotting assay were carried out to detect the protein localization and the change of expression of MCP-1 and its receptor CCR2 for comparing their difference separately at each time points.Results:As compared with the control group, there was a significant decrease of mRNA and protein expression of MCP-1 and its receptor CCR2 staining in renal fibrosis. Degree of renal injury in patients with the continuous increase, the expression level on the rise. But there is no change in control group. There were significantly difference among pathology grades (P<0.05). It was positive correlated closely with the level of fibroblast reaction phase trends. The distribution and dense parts of their positive cells is the same region.Conclusion:It is found that MCP-1 and its receptor CCR2 played a key role as an important inflammatory mediator in the occurrence and development of hydronephrosis by monitoring the expression levels of them in renal fibrosis tissue in patients with hydronephrosis. MCP-I and its receptor CCR2 may induce the reaction mechanisms involved in the occurrence of hydronephrosis by regulating fibroblasts. So we can explore some generally accepted and effective and simple means and objective marker to evaluate damage degree of hydronephrotic kidneys in patients.
