Radix Clematidis is used for analgesic;anti-inflammatory;gallbladder and antibacterial as one of Traditional Chinese Medicine from ancient times. There are many pubulished reports and literatures which studied on acient medical documents, species identification, chemical components and pharmacology about Radix Clematidis.Through collection and identification of Radix Clematidis samples from Bozhou,Yuzhou,Zhengzhou,Lanzhou and Chengdu medical materials markets,an investigation of common commercial Radix Clematidis were made out and the original plant species were clarified because of the chaos of Commercial Radix Clematidis. Clematis chinensis Osbeck and C. terniflora var. mandshurica (Rupr) Ohwi were found.Total saponins from Clematis hexapetala are attented widely as the active chemical components which had good pharmacological effects. The extracting conditions and parameters of total saponins from Clematis hexapetala by ultrasonic method was based on the single factor orthogonal test. The orthogonal test demonstrated:the sequence of the affected extracting saponins from the strongest to the weakest was the ratio of solid to liquid,extraction time,ultrasonic power and extracting temperature.The results indicated that the best extracting parameters were as follows:the concentration of alcohol was 60;the extractive times were 2;the ratio of solid to liquid was 1:20;the ultrasonic assisting time was 20min;the ultrasonic power was 360W;the extracting temperature was 70℃.The extraction rate of total saponins with alkaline extraction method was higher than that with neutral distilled water extraction method.The pharmacodynamic study in root of Clematis hexapetala Pall. produced in Gansu province which was recorded in Chinese pharmacopoeia was conducted. Dimethyl benzene (DB)-induced mouse ear edema model were employed for the anti-inflammatory activity experiment.The experiment for analgesic activity was done using acetic acid-induced writhing method.The results showed that total saponins from Clematis hexapetala at 0.034 mg/g and 0.068mg/g showed an inhibitory effect against DB-induced mouse ear edema volume and the effect of reducing the number of acetic acid-induced writhing. But the activity was not significant. The result suggests that total saponins from Clematis hexapetala has some anti-inflammatory and analgesic activity. This article has studied the effect on antitumor in vitro of total saponins. The growth inhibition was analysed by MTT in the tumor of hepatocellular carcinoma Hep-G2,hepatocellular carcinoma SMMC-7721 and cervixuteri Hela. The total saponins were found to be active in effect on antitumours in vitro.The concentration was lmg/ml,the ratio of inhibition of 3 types of tumor cells growth were above 80%, and the IC50 were 0.523mg/ml,0.195mg/ml,0.290mg/ml. The ratio of inhibition of cells has remarkable dose-dependence.This study would clarify the pharmacological activity of Clematis hexapetala and provide the theoretical basis for the development of new antitumor drugs.The Study on extraction technology and pharmacodynamics of total saponins of Clematis hexapetala Pall. produced in Gansu province in this article were reported first.
