Our study is about effect and repair of Polysaccharide from compound QiLing medicament on acute liver injury caused by CCl4 in Mice,to discusss effect mechanism of Polysaccharide from compound QiLing medicamentUltrasonic-assisted extraction was adopted to extract polysaccharide from compound QiLing medicament. The content of polysaccharide was determined by the spectrophotometer using sulphuric acid anthrone method. The optimal conditions were determined with the orthogonal design of L9(34), namely, time of 60min,temperature of 80℃, and the ratio of solid to liquid 1:10, power of 300W.0.3%CCl4 liquid paraffin solution was applied to establish experimental liver injury mode in mices, and the liver injury mices were gastrogavage administered with different doses QiLing polysaccharide(doses of QiLing polysaccharide were125mg·kg-1,250mg·kg-1,500mg·kg-1). Firstly, liver and spleen index,the activity of serum aminotransferas,the content of total bilimbln (TBiL),antioxidation index of serum,the content of nitrogen monoxidum (NO) and the activity of nitric-oxide synthase(NOS) were detected. Then, the microstructure and ultras -tructure changes of liver tissue were observed. Through all the experiments above, the effects of QiLing polysaccharide on liver injury mice were studied systematically.(1) The mice were administrated orally with QiLing polysaccharide two weeks, QiLing poly -saccharide could decrease liver and spleen index, The high dose group was notably different to CCl4 model group(P<0.05), and decrease the content of ALT,AST and TBiL in serum, The middle dose group and high dose group were very notably different to CCl4 model group(P<0.01). The mice were administrated orally with QiLing polysaccharide four weeks,the effects of each QiLing polysaccharide dose group were all obviously.(2) The mice were administrated orally with QiLing polysaccharide two weeks, QiLing polysaccharide could increase the activity of SOD in serum and decrease the content of MDA in liver homogenate. The effects of each QiLing polysaccharide dose group were very notably different to CCl4 model group(P<0.01), and increase the content of NO and the activity of NOS, The high dose group was very notably different to CCl4 model group(P<0.01). The mice were administrated orally with QiLing polysaccharide four weeks, the effects of each QiLing polysacc -haride dose group were all followed obviously. The high dose group was best of all , that wes very notably different to CCl4 model group(P<0.01).(3) The mice were administrated orally with QiLing polysaccharide two weeks, Microstructure of liver of tested mice were observed which showed that a large aera of necrosis in the centeral of the centeral veins and hepatic lobules in CCl4 model group, wide moderate and severe hydropic degeneration or ballooning degeneration and fatty degeneration, Nucleus disappears or pyknosis and red-dyed. The safeguard effects of each QiLing polysaccharide dose group were all obviously.The high dose group was best of all. The mice were administrated orally with QiLing polysacch a -ride four weeks, the repari effects of each QiLing polysaccharide dose group were all followed obviously.(4) Ultramicrostructure of hepatic of tested mice were observed which showed that the CCl4 model group of Hepatocytes organelles were dissolved or remaining a small number of organelles, mitochondrial swelling, mitochondrial ridges were lossen and breakage, The smooth endopla -smareticulum (SER) expanded severly, a large number of vacuole degeneration and fatty degeneration in the cytoplasm. The safeguard effects of each QiLing polysaccharide dose group were all obviously for hepatocyte damnifications.The high dose group was best of all.
