| Objective Through detecting the expression of MMP-10,MMP-11,TIMP-2 and DNA mutation of TIMP-2 in gastric carcinoma to disscuss the relationship between their expressions and clincopathological characteristics,and explore the machinery of molecular biology for gastric carcinoma.Methods①54 specimens resected from patients with gastric carcinoma and 20 normal gastric tissues were studied by immunohistochemical PV9000 method for the expressions of MMP-10, MMP-11 and TIMP-2.②TIMP-2 gene mutations in exon2 were examined in these 54 specimens of gastric carcinoma using PCR-SSCP analyses,and the positive specimens were confirmed by DNA sequencing.③The immunohistochemical results were analyzed using the software of SPSS13.0 by means of Rank sum test and Spearman rank correlation analysis.Results①The differences of expression of MMP-10, MMP-11 and TIMP-2 were significant between cancer mucosa and non-cancer mucosa(u=244.500～255.000, P<0.01).②Expressions of MMP-10 were correlated with tumor size,depth of tumor invasion,LN mestasis and TNM stage (X2/u=9.380～201.500, P<0.01);Expressions of MMP-11 were correlated with tumor size, depth of tumor invasion, differentiation degree,LN mestasis and TNM stage (X2/u=7.961～173.000, P<0.05);Expression of TIMP-2 was correlated with LN mestasis and TNM stage (u=192.500,224.000,P<0.05).③Significant correlation had been found between MMP-10,MMP-11 and TIMP-2 (r=0.456,0.503,P<0.01).No significant correlation had been found between MMP-10 and MMP-11 (r=0.231, P>0.05)④PCR-SSCP and silver staining displayed that TIMP-2 exon2 in all the 54 gastric carcinoma had no mutations.Conclusions MMP-10,MMP-11 and TIMP-2 expressed negatively or poorly in normal gastric tissue, but intensively in gastric carcinoma.The difference was significant.The results indicated that the abnomal expression of MMP-10,MMP-11 and TIMP-2 were related with gastric carcinoma growth.. Expressions of MMP-10 and MMP-llwere both correlated with tumor size, depth of tumor invasion,LN mestasis and TNM stage, The results indicated that they were closely related with progression and heterogeneity of gastric carcinoma.Expression of TIMP-2 was correlated with LN mestasis and TNM stage, and the positive correlation between MMP-10,MMP-11 and TIMP-2, it proved that TIMP-2 was closely related with invasion and mestasis of gastric carcinoma. We had not detected any proof to indicate correlation of the gene mutation and the higher expression of TIMP-2. We infered that the insufficinet compensation of MMPs/TIMPs leaded to high expression level of TIMP-2 in gastric carcinoma.
