Objective:Observe inflammatory cells, granulation tissue hyperplasia, and expression of a-Smooth Muscle Actin(a-SMA), transforming growth factor-β1(TGF-β1) which closely related to fibroblast proliferation, collagen activities in different periods of the repair of injuried bile duct in dogs after the treatment with Salvia, to Study the value on Salvia applicating in the prevention of injuried.Methods:66 local healthy hybrid dogs were randomly divided into control group (n=6), injuried control group (n=30), the experimental group (n=30). The establishment of animal models:normal control group be directly killed and take out the normal bile duct; the control group dogs is breeded after the manufacture of bile duct injury model in dogs; the experimental group is application of injection parameters Dan powder solution during and after the manufacturing model of bile duct injury. each group were killed 6 dogs at each time point:3d, 1wk,2wk, 1mo,3mo and take out samples of injuried bile duct. Specimens were fixed in 10% formaldehyde, paraffin-embedded after routine processing, tissue sections. HE staining, and 3 groups of specimens were observed infiltration of inflammatory cells and granulation tissue hyperplasia; by immunohistochemistry SP method (streptavidin-perosidase) in Detection of a-SMA and TGF-β1 expression of strength.Results:The normal control group, no inflammatory reaction and granulation tissue proliferation. In the early postoperative period (<2wk):the experimental group can be seen a large number of lymphocytes, macrophage infiltration,and tissue necrosis is not obvious; injuried control group can be seen the infiltration of inflammatory cells neutrophil, eosinophil white blood cells, numbering a few lymphocytes, tissue necrosis is evident. There was significant difference in inflammatory cells and necrosis,compare the the normal control group with the experimental group and the damage control group (P <0.05); The capillary proliferation is more apparent compare the experimental group with the damage control group,and there were significant differences (P<0.05). There are a small amount of expression of a-SMA and TGF-β1 in the normal control group, and the expression of a-SMA and TGF-β1 in the experimental group and the damage control group was stronger than the normal control group, there were significant differences (P <0.05). The expression of the experimental group was stronger than the injury group on the early time(P<0.05), and it was weaker at later (P<0.05). Conclusion:Bile duct injury 3d-3mo, bile duct repair is completed with biological effects such as intervention, induction, regulation, promotion of a-SMA and TGF-β1. Salvia can enhance the wound tissue inflammation, can promote angiogenesis, the formation of granulation tissue, showing the role of immune regulation, and have positive role for a-SMA and TGF-β1 in the bile injuried duct early repair; in the bile duct repair later, regulating a-SMA and low expression of TGF-β1.
