Objectives:The study is to explore the repair of skin photoaging by Dermal multipotential stem cells (DMSCs) and provide a novel route for the treatment of skin photoaging. Methods:57SD rats were randomly divided into normal group for 8 rats and model group for 49 rats, Use of UVA and UVB ultraviolet light irradiated on the model at the same time (2 h per day, continuous exposure to 60d),8 rats were randomly carried on skin photoaging for model identify.The model were randomly divided into control group for 16 rats (PBS replace) and treatment group for 25 rats(1×106 cell suspension liquid). The DMSCs of suckling SD rats were isolated and cultured in vitro, then after identifying, labeled with DAPI and took trement group for 9 rats, Several transplated in derms at skin photoaging,after transplated 1d,3d and 7d then separately put to death,and used fluorescence microscope to observe DMSC which marked DAPI in skin photoaging area of survival,migration and settlement. Then carried on cell tranplation on the rest of treatment group of rats kin photoaging. After 4 weeks,8 weeks were used immunohistochemistry, VG and Weigert stain to observe collagen fibers and elastic fibers changes in dermal fibroblasts. Results:During different time, we observed DMSCs which marked DAPI survive, diffusion, migration. Immunohistochemistry showed:Compared to control group:Fibroblasts increased, especially in superficial dermal layers in treatment group after 4 weeks.There had statistically significant differences between the two results compared (P<0.01), after 8 weeks that were widely and evenly diffused in each layer of skin, There had statistically significant differences between the two results compared (P<0.01). VG stain showed: Compared to trement group:After 4 weeks, the amount of collagen fibers which degenerated or massive degenerated had reduced, and saw a small amount of new formation of collagen fibers. After 8 weeks, the degenerated of collagen fibers that extended to subcutaneous tissue and deeply dermal layers almost disappeared, and appeared a lot of new collagen fibers which hyperchromatic and orderly, and skin structure hierarchy was clear, so had not obviously changes compared to normal group. Weigert stain showed:Compared to control group:After 4 weeks, the degenerated, gathered and tangled of collagen fibers in deeply dermal layers in treament group had obviously reduced; after 8 weeks, elastic fibers in dermal layers had almostly vanished, there were new wavy elastic fibers around hair follicle and in superficial dermal layers, so had not obviously changes compared to normal group. Conclusion:DMSCs could obviously cure skin photoaging.
