Objectives:To explore the repair of skin photoaging by dermal multipotential stem cells and provide a novel route for the treatment of skin photoaging. Methods: 48 SD rats were randomly divided into normal group for 8 rats and model group for 40 rats. Using UVA and UVB ultraviolet light irradiated on the model at the same time (2h per day, continuous exposure to 60d),8 rats with HE staining were randomly carried on skin photoaging for model identification.The model were randomly divided into treatment group for 16 rats (PBS instead) and control group for 25 rats(1×106 cell suspension liquid). The DMSCs of suckling SD rats were isolated and cultured in vitro. Then carried on cell tranplation every two weeks on treatment group of rats skin photoaging. Draw the skins from normal control group, treatment group at 4 weeks,8 weeks the treatment group to indentify the expression of P53 and PCNA in the epidermis of the rats with Immunohistochemical staining.Results:The immuno-histochemistry showed:Compared to control group:there were no obviously differents in treatment group after 4th weeks on P53. there were no statistically significant differences between the two results compared (P>0.05), The P53 are reduce in treatment group after 8th weeks that There had statistically significant differences between the two results compared (P<0.05). Compared to control group:4th weeks of treatment group and 8th weeks of PCNA skin were positive, express respectively were statistically significant comparative (P< 0.05). Compared to normal group:after 8th weeks of treatment group P53 and PCNA had no statistical difference(P>0.05). Conclusion:DMSCs could obviously cure skin photoaging in rat's skin.May be it is a new treatment to cure skin potoaging in human.
