Hepatitis C virus (HCV) is a kind of yellow fever virus, after the human infected, it can cause abnormal immune response, leading to tissue damage, forming chronic persistent infection, and to the further development of liver cirrhosis or even hepatocellular carcinoma. HCV was highly variable throughout the genome ,the 27 amino acid which in the HCV envelope protein E2 N terminal is the most, so it is named for the first hypervariable region (HVR1).The antibody test of Hepatitis C virus plays an important role in blood screening and disease diagnosis. However, because of the high HCV mutation, the current existence of HCV antibody detection reagent missed, neutralizing antibodies is difficult to detect . Institute of preclinical medicine, The Academy of military medical sciences Military had selected four high-HVR1 cross-reactive antigens, use their own carriers pBVIL1 to express these HVR1 chimeric antigen, obtained a chimeric antigen F4HVR1 which can cover most of the HVR1 antigenic variation (Patent No.: ZL 0114187916).In this study, we research on the basis of chimeric antigen F4HVR1,the chimeric antigen was purified, to identify the activity and specificity of chimeric antigen,and establish the ELISA detection technology of the HVR1 antibody.In this study, ALT and hepatitis B surface antigen (HBsAg) testing results, among 77991 blood donors in Beijing Red Cross Blood Center between 2008-2009,were statistically analyzed to study the the gender, age, seasonal distribution characteristics among donors with abnormal ALT, and also do correlation analysis between abnormal ALT and HBsAg.In this study, 2035 blood samples with high ALT level were tested for HVR1 antibodies, they were detected by F4HVRl antigen, and to compared with Antibodies against HCV Core,NS3,NS4,NS5. To find the sample only positive for HVR1,and to evaluate the important role of HVR1 antibodies testing in blood screening.We established HVR1 antibody detection reagents for the evaluation of donors with blood quality control materials. The chimeric antigen is completely different from the HCV diagnostic reagents currently used, and the chimeric antigen improves the sensitivity of HCV screening.
