Objective:Hepatitis B virus (HBV) is the most important pathogenic factor for hepatocellular carcinoma. A number of patients infect HBV in a wide range of China.HBV DNA,alanine aminotransferase enzyme (ALT),aspartate aminotransferase (AST) and hepatitis B three system are usually used for monitoring HBV replication and estimating liver injure,but those markers are all have limitation.miR-122is a liver-specific microRNA,it is not only involved in various physiological processes in hepatic such as growth,differentiation and so on,but also in pathology.Here,we explored miR-122levels in different groups,and investigated whether miR-122levels correlate with viral replication.Furthermore,we examined the relation among miR-122, ALT,AST,and whether miR-122may be a new marker for liver injure.Methods:80cases were collected,including60cases of hepatitis B,which were stratified that depended on HBV DNA load.and there were20healthy controls. The concentration of miR-122was assessed by quantitative real-time reverse-transcription PCR.ALT,AST were detected by automatic biochemical analyzer.2△△CT method was used for HBV infectived comparing with healthy controls.2△CT method was used for statistics.Results:1.miR-122,ALT,AST are all elevated in groups of hepatitis B surface antigen-positive.miR-122was25.16times in HBV SAg+than HBV SAg-.The mean ranks of ALT,AST in HBV SAg+were45.29,47.84,and in HBV SAg-were26.13,18.48,P<0.01.2.Cases of HBV SAg+were stratified into three groups which denpended on HBV DNA load,miR-122was higher in7-8power of HBV replication than other groups,P<0.05. miR-122in2-3power group compared with no replication group,there was no statistically significant,and it compared with healthy controls,p=0.04.there also was no statistically significant between no replication group and healthy controls.3.ALT,AST in groups of HBV SAg+were no significant difference,P>0.05.compare ALT,AST in groups of HBV SAg+and healthy controls,ALT:P=0.003,P=0.009,P=0.033,AST:P≤0.01AThe correlation coefficient between miR-122with ALT was0.251, P=0.025. with AST, the correlation coefficient was0.379, P=0.001.the correlation coefficient between AST with ALT was0.821, P<0.01.Conclusion:1.miR-122is elevated in hepatitis B,and it is positive correlation with HBV DNA replication.2.miR-122is correlate with ALT,AST,and it is sensitive for liver injure.3.miR-122is a better liver injure marker than ALT、AST.4.miR-122in plasma is a new marker for liver injure,although it is not obviously increased in low HBV replication.lt maybe caused by immune dysfunction and HBV virus activity.lt also need more samples in further study.
