Objective:The kansui roots were Euphorbia kansui T. N. Liou ex T. P. Wang, belongs to Euphorbiaceae Euphorbia kansui. As known as "offal good medicine" by ancient medical scientists, Euphorbia kansui was a traditional medicine for treatment of drastic purgation and hydragogue.Moreover kansui possessed therapeutic effect such as watery diarrhea, breaking accumulation,open urine and stool. In clinical application, kansui was used for edema turgor, thorax hydrocele, retention of phlegm and fluids accumulation, abdominal effusion, abnormal rising of vital energy and dyspnea with cough, bowel and bladder dysfunction, detumescence and eliminating stagnation. Pyogenic infection with abscess used for external application. The property of kansui was bitter and cold,moreover had rank poison. It may stimulate skin and mucous membranes effect, and overdose will appear abdominal pain, vomiting and diarrhea,dehydration,severe dyspnea circulatory failure and death. Therefore,using processing kansui root in order to reduce toxicity and alleviate drug property to guarantee the safety in clinical application. This topic was mainly study on whole chemical constituent changes of the raw kansui root and kansui root by vinegar. The result showed that the relationship between potency and toxicity,as well as come to elaborated the mechanism of processing drug effect attenuation and synergia. Supported by the scientific and technological project in Hubei Province (2007AA402A43). Methods and Results:1. According to the difference of polar part, study the chemical constituent of raw kansui root and kansui root by vinegar, with solent extraction method which divided into aether petrolei extract, chloroform extract, acetic ether extract and water extract.Using high performance liquid chromatography to analysis the different polar part of the whole changes in chemical composition.2. Used mice as target subjects, to experimental raw and kansui root by vinegar extracts with adopt the small intestine propulsion and acute toxicological experiment. The result showed that processing kansui root's aether petrolei extract, chloroform extract, acetate ether extract than raw kansui root, the cathartic effect has alleviated and the toxicity has weakened.3. With solvent and chromatography method to purified chloroform extracts of kansui root by vinegar.Apply gel, ToyopearlHW-40, SephadexLH-20, column chromatography to enrichment kansui chromatog-raphic fraction. Three compounds were isolated and two compounds were identified.Conclusion:The topic crossed multidisciplinary research methods of Chinese Chemical, Chinese Pharmacology and Analytical Chemistry etc., studied pharmacological effects and the chemical constituents of the raw kansui root and kansui root by vinegar. The result showed that the aether petrolei extract, chloroform extract and acetate ether extract of raw kansui root and kansui root by vinegar were not only the effectiveness cathartic effect but also the toxicity part.After processing by vinegar, the raw kansui root chemical contents had certain quantitative and qualitative change,and palliationed the potency and toxicity. Preliminary studied the mechanism of attenuation processing raw kansui root by vinegar.
