Objective:In this study,the toxicity of different extracts of radix glycyrrhizae was investigated.The pyrolysis and combustion characteristics of different extracted parts of radix glycyrrhizae were analyzed by using thermal analysis technique,and the optimum temperature of processing was obtained.The processing technology of radix glycyrrhizae was optimized by response surface test.With the aid of diuretic experiment,acute toxicity experiment and intestinal toxicity experiment,the effects and toxicity changes of gansui vinegar were studied before and after broil,and the mechanism of processing and detoxification of gansui was preliminarily explored from the levels of anti-inflammatory factors and proinflammatory factors.Methods:1.Using petroleum ether,chloroform,ethyl acetate and n-butanol as the study subjects,cck-8 method and LDH activity detection method were used to compare the toxicity of different extracts of radix glycyrrhizae on small intestinal epithelial cells?iec-6cells?of rats,and to screen the toxic sites.2.The pyrolysis and combustion characteristics of different parts of radix glycyrrhizae powder,radix glycyrrhizae control material powder and radix glycyrrhizae were studied by means of thermal analysis technology.3.The effects of adding amount of vinegar,frying temperature and frying time on processing technology of glycyrrhizoma glycyrrhizae were investigated by using response surface test with three factors and three levels.4.A water load model was established to compare the diuretic effects of different processed products of gansui on water load mice.5.LD50values of radix glycyrrhizae and radix glycyrrhizae were obtained in ICR mice on the basis of acute toxicity test.The effects of glycyrrhizoma glycyrrhizae and glycyrrhizae acetate on gastrointestinal toxicity in mice were studied by using appropriate concentrations of the drugs.6.Wistar rats were selected as the study objects,and the processing mechanism of processed glycyrrhizae glycyrrhizae was preliminarily studied by comparing the pathological damage of glycyrrhizae glycyrrhizae and processed glycyrrhizae glycyrrhizae on the gastrointestinal tract of rats,measuring the liver,spleen and kidney indexes,as well as the levels of IL-1,IL-6,TNF-?,IL-4 and IL-10 in the serum of rats.Results:1.The proliferation-toxicity test results of cck-8 cells were consistent with the LDH activity test results,suggesting that radix glycyrrhizae had obvious enterotoxicity,and the toxicity levels of each site were respectively petroleum ether site,chloroform site,ethyl acetate site and n-butanol site.2.According to different raw euphorbia kansui pyrolysis combustion characteristic of different extraction parts,deduce the euphorbia kansui vinegar main point of the best processing temperature is 276.4?,the maximum temperature of 329.4?.3.The optimum processing conditions of vinegar broiled euphorbia kansui is:put a proper amount of raw euphorbia kansui,add rice vinegar 30%,at270?under making time is 3.5 min.4.In lavage after dosing for 1 to 2 hours,positive and various dosage groups have obvious diuretic effect,after dosing for 3 to 6 hours,positive life euphorbia kansui,high-dose group,vinegar euphorbia kansui high dose group showed strong diuretic effect,compared with the model group with very significant difference?P<0.01?,namely,positive medicine within 3 h after dosing diuretic effect is stronger,3 hours to 6 hours gradually;The diuretic effect was increased between 3 hours and 6 hours in the high dose group and the high dose group,suggesting that the diuretic effect of high dose of glycyrrhizoma glycyrrhizae was lasting.5.According to the results of the acute toxicity test,the LD500 of shenggansui was 77.91±5.88g/kg,with a 95%confidence interval of79.469-103.80g/kg;the LD500 of acetoprene was 129.89±3.25g/kg,with a 95%confidence interval of 112.88-149.49g/kg.It can be seen from the experiments of gastric emptying and intestinal propulsion that there was a significant difference between the groups with different doses of shenggansui and the blank control group?P<0.01?,and the intestinal propulsion effect of the groups with different doses of shenggansui and the group with different doses of acetanilide on mice was significantly higher than that of the control group.In addition,the gastric residual rate of the other groups except shenggao group was significantly lower than that of the blank group?P<0.01?,indicating that different processed products of gansui could promote gastric emptying.There were significant differences between the two groups?P<0.05,P<0.01?.6.Through the pathological examination of the duodenum and jejunum of rats,it was found that the gansui product group showed different degrees of pathological damage,including edema,hyperplasia,necrosis and shedding of mucosal villous epithelial cells,etc.,and the stimulation effect became more significant with the increase of dose between the groups.Among the processed products,vinegar processed products have a less stimulating effect on the intestinal tract.Born at the same time,euphorbia kansui product the liver index and spleen index of each dose group compared with the blank control group,all have significant difference?P<0.01?,and have high group,compared to the liver index and spleen index of vinegar in the low group,vinegar,vinegar high group have significant difference,that euphorbia kansui vinegar after acupuncture is lowering the toxicity of the rat liver toxicity,spleen.However,medium and high doses of radix glycyrrhizae and radix glycyrrhizae acetate had a certain effect on kidney injury in rats.The high dose group of radix glycyrrhizae acetate had the strongest effect on kidney injury,while the low dose group had no significant difference from the blank group.The low dose of radix glycyrrhizae acetate had no toxicity on kidney.According to the detection results of several cytokines,the content of anti-inflammatory factors IL-4 and IL-10 in processed glycyrrhizoma glycyrrhizae was higher than that in unprocessed glycyrrhizae glycyrrhizae,and the levels of tumor necrosis factor-?,proinflammatory factor IL-6 and IL-1?were relatively lower,indicating that the content of proinflammatory factors and anti-inflammatory factors in serum of glycyrrhizae glycyrrhizae rats was close to a dynamic balance.Conclusion:This study on the basis of the known toxicity of euphorbia kansui parts,using thermal analysis technology analysis of euphorbia kansui pyrolysis characteristics of different extraction parts,get euphorbia kansui vinegar main point of optimum temperature of 276.4?.As a reference,using the response surface method to optimize the process parameters of euphorbia kansui vinegar,namely content of quantity of vinegar was 30%,making 3.5 min under 270?.It was found in the comparison of the pharmacodynamics and toxicity of glycyrrhizae glycyrrhizae with the best processing technology that glycyrrhizae glycyrrhizae had the effect of reducing toxicity and preserving efficacy.The serum levels of anti-inflammatory factors in the treated rats were relatively increased,which promoted the dynamic balance between anti-inflammatory factors and proinflammatory factors,providing certain reference for the processing mechanism of reducing toxicity of glycyrrhizae glycyrrhizae.Meanwhile,from process optimization,quality evaluation to efficacy verification,it provides support for the clinical application of glycyrrhizae.
