Purpose This topic want to detect Livin gene expression in the peripheral blood of patients with colorectal cancer, to analyze its correlation with clinical stage and pathological. To explore whether Livin mRNA can became a tumor marker of colorectal cancer. By detecting Livin gene expression changes of preoperative and postoperative colorectal cancer patients to infer the effect of laparoscopic surgery and open surgery to micro-metastasis of postoperative malignant tumor.Methods64patients with colorectal cancer were randomly divided into laparoscopic group and open group,32cases in each group. Use RFQ-PCR to detect all patients’relative quantity of peripheral blood at pre-operation and1day after operation and7days after operation. Choose20cases of colorectal benign tumor and20cases of healthy adults as controls, to detect the expression of peripheral blood Livin mRNA by RFQ-PCR.Results1. The ACt of the group of64cases colorectal cancer patients was5.83±1.37. The ACt of the group of20cases colorectal benign tumor was8.96±1.46. the ACT of healthy control group was9.87±1.25. Use healthy group As a control, the-AACt of the benign tumor group was0.91±1.89, the-AACt of colorectal cancer group was4.04±1.80.there was significant difference between the colorectal cancer group and colorectal benign tumor group (P<0.05). But no significant difference between the colorectal benign tumor group and the healthy control group (P>0.05). The average relative quantity of the colorectal benign tumor group and colorectal cancer group were1.87and16.45times as the normal control group.2. Colorectal cancer patients were divided into groups by Dukes stage. There were23cases in Dukes B and36cases in Dukes C and5cases in Dukes D. The-AACt of the Dukes B group and the Dukes C group and the Dukes D group were3.51±1.83and4.09±2.02and6.31±1.49. The difference was no statistical significances between the Dukes B group and the Dukes C group (P>0.05). Compared with the Dukes B group and the Dukes C group, the Dukes D group had significant difference (P<0.05). The mean relative quantity of the Dukes B group and the Dukes C group and the Dukes D group were11.39and17.03and79.34. There was a obvious rising trend.3. Grouped by pathologic differentiation degree,26patients were well-differentiated group,32patients were moderately differentiated group,6patients were poorly differentiated and undifferentiated group. The-AACt of well-differentiated group and moderately differentiated group and poorly differentiated/undifferentiated group were3.21±1.68and4.06±1.76and6.25±1.67. There was no significant difference between moderately differentiated group and well-differentiated group (P>0.05). There was statistical significance between Poorly-differentiated/undifferentiated group and well-differentiated group (P<0.05). The mean relative quantity of well-differentiated group and moderately differentiated group and poorly differentiated/undifferentiated group were9.25and16.68and76.11. There showed a clear rising trend.4. The-AACt of laparoscopic surgery group before operation and1day after operation and7days after operation were4.06±1.69and3.90±1.61and0.76±1.89, The-AACt of the open surgery group before operation and1day after operation and7days after operation were4.03±1.67and3.86=1=1.63and0.66±1.86. There were no statistically significant difference between the two groups compared in each time point (P>0.05), also no difference between the before operation and1day after operation in each group (P>0.05). But there were significant difference compared7-days after-operation with before-operation and1-day-after-operation in each group (P<0.05).Conclusions The mean relative quantity of Livin gene in peripheral blood of patients with colorectal cancer was significantly higher than the healthy control group and the colorectal benign tumor group. Livin mRNA is expected to become a tumor marker for colorectal cancer after further quantitatively research by expand the sample.The mean relative quantity of Livin gene in peripheral blood of patients with colorectal cancer was positively correlated with its Dukes stage. The mean relative quantity of Livin gene in peripheral blood of patients with colorectal cancer was negatively correlated with the degree of pathologic differentiation.The changing trends of the mean relative quantity of Livin gene in peripheral blood between laparoscopic surgery and open surgery patients were similar. Suggested the Laparoscopic surgery does not increase the risk of blood metastasis of colorectal cancer.In this experiment, the numbers of samples were small. So the conclusions may have certain deficiencies. Needs further study by expand sample numbers.
