After the technique, rabbits were randomly divided into 1%Matrine drop eye fluid experimental group, 0.1%DXM positive control group, normal saline negative control group, each group of 21.apply drugs clinically for 3 months continuously to rabbits by simulating. 1week, 1month, 3 months after PRK,the rabbits were sacrificed in batches. Observe rabbits eye in the Slit-lamp microscope and record the Haze classification conditions. Rabbit corneas is made into the paraffins specimen. The slices were done HE dyeing , collagen dyeing examination ,checking the expression of TGF-β1,α-SMA positive cells of the rabbit anterior corneal stromal in immunohistochemical detection and counting the number ofα-SMA positive cells.Result:1.After PRK, observes the Haze situation in the slit-lamp microscope which 1% Matrine group and 0.1% DXM group was significantly lighter than the saline group(P<0.05).1% Matrine group and 0.1% DXM group Haze classification have no difference(P>0.05).2.The number of the anterior cornea stromal mean proliferation levels of new collagen in 1% Matrine group and 0.1% DXM group less than those in saline group. proliferate active low.1% Matrine group has no difference comparing with 0.1% DXM group.3.TGF-β1 expression of 1% Matrine group and 0.1% DXM group in the cornea were significantly weaker than the NS group, the difference with a significant (P<0.05), expression of TGF-β1 in 1% Matrine group and 0.1% DXM group is almost no differences (P> 0.05). 4.α-SMA positive cells in 1% Matrine group and 0.1% DXM group of corneal were significantly less than the NS group, the difference with a significant (P<0.05), andα-SMA-positive cell count in 1% Mat group and 0.1% DXM group is basically no difference (P> 0.05).5.α-SMA positive cell counts and TGF-β1 expression level in corneal stroma after PRK in rabbit eyes is linear positive correlation between the two.(r = 0.949, P<0.01).Conclusion:1. 1% Matrine eye drops can effectively inhibit the formation of Haze after PRK in rabbit eyes. 2. The possible mechanism that Matrine inhibits the formation of Haze is to inhibit fibrolast proliferation and new collagen formation in the corneal stroma after PRK in rabbit. Reduce expression of TGF-β1 andα-SMA.
