The Relation Of Angiopoietin-2 With Mvd And Mlvd In Colorectal Adenocarcinoma

Objective: We recorded the expression of angiopoietin-2(ANG-2)mRNA,analyzed the relationship with clinical pathology of colorectal adenocarcinoma(CRC).We explored the characteristics of form and distributionthe in the CRC's microvascular and lymphatic ; counted the microvesseldensity(MVD) and microlymphatic vessel density(MLVD) in CRC also.we investigated the correlation with the expression of ANG-2mRNA with angiogenesis and lymphangiogenesis in CRC indirectly.Methods: Collecting 50 newly diagnosed cases of CRC and 12 cases of benign lesion pathology specimens of the First Affiliated Hospital of Nanhua University from July 2009 to November 2009 , applied RT-PCR detect the expression level of ANG-2mRNA in cancer tissue,benign tissue and normal surgical margin tissue; we analysed MVD,MLVD by morphometric which marked by CD34,LYVE-1 respectively,and we analyzed the relationship between MVD,MLVD with clinical pathology of CRC.We also assayed the relationship between the expression of ANG-2mRNA with MVD,MLVD in CRC.Results: (1)In all the samples,the positive rates of ANG-2mRNA in CRC's cancer tissue,benign tissue and normal surgical margin tissue were (32/50,64.0%),(1/12,8.3%)and(3/50,6.0%)respectively.(2)There were 32 samples that the ANG-2mRNA were positive,and among these samples the intratumoral MVD and peritumoral MLVD were higher than that in negative group of CRC (P<0.01;P<0.01);there were a strong correlation between the ANG-2mRNA expression level and intratumoral MVD,peritumoral MLVD respectivly (p<0.05,r=0.704;p<0.05,r=0.493).(3) The CRC's intratumoral and peritumoral MVD were higher than the normal surgical margin's (P<0.01),the intratumoral MVD was higher than the peritumoral MVD additional(P<0.01).(4)The MLVD of the peritumoral and the normal surgeral margin's were higher than the intratumoral MLVD in CRC(P<0.01),the peritumoral MLVD was higher than the normal surgeral margin's MLVD additional(P<0.01).(5)The level of intratumoral MVD and peritumoral MLVD in Duke's A/B stages was lower than C/D stages of CRC(P<0.05),and it has significant relationship with the distant metastasis;the MLVD with lymphatic metastasis were higher than without lymphatic metastasis(P<0.05),but it has no relation with the MVD(P>0.05).Conclusions : (1)ANG-2 was high expressed in CRC,the ANG-2 expression levels have closely relation with intratumoral MLD and peritumoral MLVD.(2)MVD was the highest in the intratumoral and have relation with Duke's stage,tumor cell differentiation and distant metastasis;MLVD was the highest in the peritumoral and have relation with Duke's stage,tumor cell differentiation and lymphatic metastasis.
