| Objective:To study the effects of the cell proliferation and its apoptosis of diallyl disulfide(DADS) on human lymphoma Raji cell. To determine if there are chemotherapy senditizing about diallyl disulfide on human lymphoma Raji cell againsted by vincristine(VCR) and its molecular mechanism.To explore the treament of lymphoma and rationl combination of drugs to provide experimental evidence.Methods:1.Raji cells were cultured in vitro. Cell proliferation inhibition of DADS and/or VCR on tumor cells were determined by CCK-8 assay.To determine the non-cytotoxic dose and VCR-IC50.2.To use non-cytotoxic dose of DADS and the VCR alone and in combination act on Raji cells.Inhition of the respective were examined by CCK-8 assay.The value of q were seeked by jin's formual.To evaluated the combined drug effects.3. The apoptosis of Raji cells were tested by flow cytometry(FCM).4.The expression of Bax and Bcl-2 in cells was assessed by immunohistochemical methods.To use the pathological image analysis software and semi-quantitative analysis to explore the DADS possible mechanisms of chemosensitivityResults:1.VCR single-agent(6.25,12.5,25.0μg/ml),DADS single-agent (0.625, 1.25μg/ml,DADS(0.625,1.25μg/ml) with VCR(6.25,12.5,25.0μg/ml) combined on the Raji cells, the growth inhibition rate at the combined group was significantly higher than the corresponding VCR single-agent group,q>1.15, DADS has a sensitizing effect.2.after 24h,VCR at different concentrations(6.25,12.5,25.0μg/ml) in single and its relationship with DADS at non-cytotoxic dose (0.625μg/ml) induced Raji cells apoptosis some extent, the apoptosis rate at the two drugs in combination were significantly higher than the corresponding single-agent group of VCR(P<0.05).3. VCR at different concentrations(6.25,12.5,25.0μg/ml) in single on Raji cells can downregulate expression of level Bcl-2 protein and upregulate expression of level Bax protein.The expression of level Bcl-2 protein were significantly lower(P<0.05) and the expression of level Bax protein were significantly higher in Raji cells at the combination of its relationship with DADS at non-cytotoxic dose(0.625μg/ml) than the corresponding single-agent group of VCR(P<0.05).The ratio of Bax/Bcl-2 increased(P<0.05).Conclusions:1.DADS at non-cytotoxic dose enhances the effect of growth inhibtion of vincristine on Raji cells since the sensitizing effect of chemotherapy.2. The effect of chemosensitization to vincristine of DADS on Raji cells may be associated with cell apoptosis.3.The effect of chemosensitization to vincristine of DADS on Raji cells may be associated with downregulating expression of level Bcl-2 protein and upregulating expression of level Bax protein. |
