Objective:To primarily investigate molecular mechanism of diallyl disulfide (DADS) stopped CNE2 cell cycles, related genes of DADS regulated cell cycles was detected by genic microarray in nasopharyngeal carcinoma CNE2 cell.Methods:MTT method detected 50% inhibiting concentration (IC50) that DADS inhibted CNE2 cells proliferation. The cell growth curve analyzed inhibitory action that DADS affected CNE2 cells growth; The cell cycle of DADS affected CNE2 cells was checked by flow cytometry (FCM). DADS regulated related genic expression of CNE2 cell cycles was detected by genic microarray. The differentially genic expression was confirmed by RT-PCR, Wester blotting and immuocytochemistry methods.Results:(1) IC50 that DADS inhibted CNE2 cells proliferation was 30 mg·L-1. With DADS concentration increased and treatment time extended, CNE2 cells proliferation was obviously inhibited by DADS (n=3, P<0.05). (2) The cells number of G0/G1 period significantly multiplicated after DADS incubated CNE2 cells. Meanwhile, cells number of S period almost no-changed. But cells number of G2/M period obviously decreased. With DADS concentration increased, cells number of G0/G1 period gradually raised (n=3, P< 0.05). (3) Genic microarray together detected 1074differential expression genes. There was 31 differential expression genes that related cell cycle (20 genes up-regulation expression and 11 genes down-regualtion expression). (4) The expression of PLK1 gene and SASH1 gene detected by genic microarray was as same as the results of RT-PCR, Western blotting and immuocytochemistry methods verification.Conclusion:1. DADS can inhibit proliferation of CNE2 cells with its concentration increased and treatment time extended which was probably related that it hold-up cell cycle of G0/G1 period.2. DADS stopped cell cycle was probably related with up-regualtion the expression of SASH1, CCNG2, DUSP1 and CDKN2B genes and with down-regualtion the expression of PLK1, GTSE1, RCC1 and ANAPC7 genes.
