Objective : To investigate and compare the expression of RHAMM gene and TPX2 gene in esophageal squamous cell carcinoma (ESCC)tissues, the para-cancer tissues and matched esophageal normal mucosa tissues,study the relationship between RHAMM gene/TPX2 gene and clinicopathological features, such as depth of invasion, histologic grade and lymph node metastasis, analyze the correlation between RHAMM mRNA and TPX2 mRNA, approach the role of RHAMM gene and TPX2 gene in tumorigenesis as well as progression in ESCC and their clinical significant.Method :Reverse transcriptase-polymerase chain reaction (RT-PCR) was used to detect the expression of RHAMM mRNA and TPX2 mRNA in 40 cases of esophageal squamous cell carcinoma tissues, the para-cancer tissues and matched esophageal normal mucosa tissues,and their relation to clinical pathologic characteristics of ESCC was analyzed.Result :1. The positive expressions of RHAMM mRNA and TPX2 mRNA in ESCC tissues(65.0% and 75.0%) were significantly higher than those in para-cancer tissues(37.5% and 47.5%) and esophageal normal mucosa tissues(22.5% and 40.0%).2. Significant relationship was observed between positive expression of RHAMM mRNA and lymphnode metastasis(P<0.01). Significant relationship was observed between positive expression of TPX2 mRNA and lymphnode metastasis or degrees of differentiation(P<0.05).3. A significant association was found between the mRNA expression of TPX2 and RHAMM(P<0.01,r=0.39).Conclusion:1.The abnormal expression of RHAMM mRNA and TPX2 mRNA were associated with the occurrence and progression of ESCC. RHAMM gene and TPX2 gene maybe act as one of biological indicators of the invasion and metastatic potential in ESCC.2. Significant association was found between the mRNA expression of RHAMM and TPX2. Detecting the expression of RHAMM gene combined with TPX2 gene may guide early diagnosis, prognosis and therapy of ESCC.
