Objective: To research the expression and corelationship of TPX2, CyclinB2and Bcl-2in esophageal squamous cell carcinoma(ESCC) for investigating their clinicalsignificance in pathogenesis and progression of ESCC.Methods:1.The genes TPX2, CyclinB2and Bcl-2were identified on gene level byReverse Transcription PCR, in18cases of ESCC and adjacent normal tissue.2. Theproteins of TPX2, CyclinB2and Bcl-2were identified in100cases of ESCC,30casesof normal esophageal epithelium,30cases of atypical hyperplasia byimmunohistochemistry, and analyzed their relationships with the clinicopatholoicalparameters of ESCC.Results:1. The genes TPX2, CyclinB2and Bcl-2were amplified by RT-PCR in18cases of adjacent normal tissue and ESCC, which of TPX2mRNA was expression levelsof0.8221±0.1236,1.5853±0.1163, significant differences were found betweenthem(P<0.01), which of CyclinB2mRNA was expression levels of0.2602±0.0712,0.3600±0.0812, significant differences were found between them(P<0.01), and whichof Bcl-2mRNA was expression levels of0.6019±0.0764,0.8666±0.0953, significantdifferences were found between them(P<0.01).2. Immunohistochemical method wasapplied to detect the expressions of TPX2, CyclinB2and Bcl-2in100cases of ESCC,30cases of normal esophageal epithelium,30cases of atypical hyperplasia; Thepositive expression rate of TPX2, CyclinB2and Bcl-2in ESCC were higher than that ofnormal esophageal epithelium and atypical hyperplasia(P<0.01,<0.05), the expressionof TPX2and Bcl-2in normal esophageal epithelium and atypical hyperplasia was nosignificant difference(P>0.05). the expression of TPX2, CyclinB2and Bcl-2in ESCCwas associated with the degree of differentiation, the lymohatic metastasis, clinicalstage(P<0.01,0.01,0.05), And there was positive correlation between TPX2withCyclinB2, TPX2with Bcl-2and CyclinB2with Bcl-2(r=0.588,P<0.01; r=0.373,P<0.01; r=0.350, P<0.01;)。Conclusion:1. The expression of the TPX2mRNA, CyclinB2mRNA and Bcl-2mRNA in ESCC were higher than in normal esophageal epithelium..2. The protein expressions of TPX2, CyclinB2and Bcl-2were highly expressed inESCC, the expression of TPX2, CyclinB2and Bcl-2in ESCC was significant differencewith the expression in normal esophageal epithelium and atypical hyperplasia,suggesting that the three proteins may be involved in ESCC development.3. The expression of TPX2, CyclinB2and Bcl-2was associated with the degree ofdifferentiation, the lymohatic metastasis, clinical stage.4. TPX2, CyclinB2and Bcl-2there was a positive correlation between them, they maybe have interaction in the occurrence of ESCC.
