Objective:To preliminary study the anti-tumor effect of dolabrane diterpenes of Ceriops tagal in Lewis Lung Carcinoma mice and its mechanism.Methods:After the establishment of subcutaneously transplanted solid tumor models with Lewis lung carcinoma cell, all the models were randomly divided into 5 groups:20,10 and 5mg/kg dolabrane diterpenes groups, saline group as tumor control group and CTX (10mg/kg) group as positive group, respectively. Then 7 days successive administration were given to all the carcinoma models and the mice were killed 24 hours after the last medication. Then the tumors of each group were weighted and the tumor inhibition rates were calculated. Also we detected the apoptosis rates using flowcytometry and the expression level of the tumor-associated transcription factor NF-KBp65 using immunohistochemistry respectively.Results:The tumor inhibition rates of the 20,10 and 5 mg/kg dolabrane diterpenes groups were 44.12%,25.74% and 22.59%. Compared to the tumor control groups, there were obvious tumor inhibition effects in dolabrane diterpenes groups and each group did not have obvious organ toxicity (P<0.05). The apoptosis rates positively correlated with the dosage of dolabrane diterpenes. And the expression level of the NF-KBp65 in the experimental group were lower than that in the control groups (P<0.05).Conclusion:the dolabrane diterpenes of Ceriops tagal has significant anti-tumor effect in the LLC mice and the mechanism may associates with the induced apoptosis of tumor cell and the lower expression levels of the tumor-associated transcription factor NF-KBp65.
