Objective:To study the anti-tumor effect of dolabrane diterpenes (Tagalsins A) of the mangrove plant Ceriops tagal and its mechanism.Methods:The subcutaneously transplanted solid tumor models with Hepatoma-22 and the ascitic tumor models with Hepatoma-22 were established. The models were divided randomly into 5 groups:5,10 and 20 mg·kg-1 Tagalsins A groups, saline group and CTX group (10 mg·kg-1), each of which was composed of 10 mice. The Tagalsins A were given by intragastric administration, the CTX by intraperitoneal injection for 7 days, same volume of saline was given to the control group. The mice were killed 24 hours after the last medication, the tumor inhibition rate, increase in life span, indexes of organ, apoptosis rate were calculated.Results:Compared with NS group, the Tagalsins A significantly inhibited the growth of Hepatoma-22 solid tumors (F=8.90, p<0.01). The tumor inhibition rate that in the 5,10 and 20 mg·kg-1 Tagalsins A groups were 24.49%,35.37%,48.30%, there were no statistically difference between the 10,20 mg·kg-1 Tagalsins A groups and CTX group (q=2.85,1.01, p>0.05). Compared with NS group, Tagalsins A slgnillcanlly prolong the survival time of H22 ascites tumor-bearing mice (F=17.06, p<0.01), reduce the thymus index (F=21.82, p<0.01), while elevated liver index of H22 solid tumor (F=121.84, p <0.01). The rate of apoptosis of tumor cells in each group of Tagalsins A were significantly higher than the NS group (F=1277.68, p<0.01).Conclusions:The Tagalsins A has significant anti-tumor efect, it's mechanism of antineoplastic action could be attributed to its action of inducing apoptosis of tumor cell.
