PURPOSE To investigate the effects of free fatty acids (FFAs) on proliferation of human periodontal ligament fibroblasts (hPDLFs) in vitro and research the possibly mechanisms.METHODS Cultured primary human periodontal ligament fibroblasts (hPDLFs) were incubated with various concentrations of Oleic acids for 24h-72h. Inhibition of proliferation was measured by colorimetric3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide (MTT) assay; then we used 0.5 mmol/L of FFAs to induced the hPDLFs, the effects of MMP-2, MMP-3 and TIMP-1 were determined by immunoflorescence and sandwich-in ELISA.RESULTS MTT assay showed that when cells were incubated in 0.5 mmol/L,the OD vaule was 0.237±0.005(P<0.01) comparing with the control group at 48h and at 72h the OD vaule was 0.237±0.005(P<0.01). FFAs inhibited the proliferation of primary human periodontal ligament fibroblasts (hPDLFs) in a dose- and time-dependent manner by MTT assay. The expression of MMP-2 and MMP-3 were 70.2±4.943 ng/ml(P<0.05),69.733±3.456ng/ml(P<0.01)at 48h and 78.6±3.081 ng/ml (P<0.01),78.067±3.421 ng/mL (P<0.01)at 72h, which increased significantly compared with the control group. The expression of TIMP-1 were 44.467±0.833 ng/mL (P<0.05)at 48h and 40.767±1.943 ng/mL (P<0.01) at 72h, which also increased significantly compared with the control group. When cells were incubated in FFAs,a significant higher expression of MMP-2,MMP-3 were observed but the expression of TIMP-1 was lower.CONCLUSION FFAs could inhibit proliferation of hPDLFs and may affect ECM metabolism depending on the functional role of MMP-2,MMP-3 in vitro, and this may imply a delayed repare of the periodontal tissue.
