Objective:To explore the expression characteristics of integrinβ1,β3 in peripheral blood lymphocytes in infertile women during peri-implantation in IVF-ET cycle and in order to provide some data for the time selection of ET as well as for judgement of treatment outcomes.Methods:①The expression level of integrinβ1,β3 mRNA in peripheral blood lymphocytes were detected by real-time quantitative fluorescence PCR at the day of hCG injection, and the ovum pick-up, the day of embryo transfer and the second, the fourth, the sixth, the eighth, respectively after the embryo transfer.②The percentage of lymphocytes expressing integrinβ1,β3 in the peripheral blood were detected by flow cytometry at point of time above-mentioned.Results:①It were progressive increase that the percentage of lymphocytes expressing integrinβ1,β3 in the peripheral blood and the expression level of integrinβ1,β3 mRNA in peripheral blood lymphocytes after the embryo transfer day in pregnancy group. It were progressive decline that the percentage of lymphocytes expressing integrinβ1,β3 in the peripheral blood and the expression level of integrinβ1,β3 mRNA in peripheral blood lymphocytes after the embryo transfer day in non-pregnancy group.②The percentage of lymphocytes expressing integrinβ3 in the peripheral blood was lower in pregnancy group compared with non-pregnancy group at the day of embryo transfer (P<0.05). The expression level of integrinβ1,β3 mRNA in peripheral blood lymphocytes was lower in pregnancy group compared with non-pregnancy group at the day of embryo transfer (P<0.05, P<0.01).③The percentage of lymphocytes expressing integrinβ1,β3 in the peripheral blood was higher in pregnancy group, compared with non-pregnancy group at the eighth day after the embryo transfer (P<0.05, P<0.01) The expression level of integrinβ1,β3 mRNA in peripheral blood lymphocytes was higher in pregnancy group compared with non-pregnancy group at the eighth day after the embryo transfer (P<0.01, P<0.01).Conclusion.:①The high expression level of integrinβ1 ,β3 on peripheral blood lymphocytes at the day of embryo transfer, may be one of the reasons of failure in IVF/ ICIS-ET cycle.②Detection the level of integrinβ1,β3 on peripheral blood lymphocytes at the day of embryo transfer, possibly as the basement for choosing the timing of embryo transfer individually in IVF/ICIS cycle.③The dynamic monitoring to the expression level of integrinβ1,β3 on PBL after the day of ET, especially the eighth day after ET, may provide a indicator for early judgement of treatment outcomes.
