Pachysandra terminalis Sieb.Zucc is widely distributed in the southwest and northwest of the People's Republic of China. As a Chinese folk medicine, Pachysandra terminalis has the properties of reducing tussis, expelling wind and removing cold, promoting blood circulation to arrest pain. Mordern pharmacology research suggests Pachysandra terminalis has the activities of cytotoxic, anti-ulcer and exhibiting anticholinesterase activity.The occurrence of steroidal alkaloids in Pachysandra is well documented, but little phytochemical work has been carried out on Pachysandra terminalis. In order to search the bioactive constituents from natural source, we began to study the chemical constituents from Pachysandra terminalis. By repeated column chromatography, including Silica gel, Toyopearl HW-40 and preparative HPLC, more than 37 compounds were isolated from liposoluble extract of Pachysandra terminalis. By using IR, MS,1H-NMR,13C-NMR,1H-1H COSY, HMBC, HSQC spectra and chemical methods, five new alkaloids, (+)-(20S)-20-(dimethylamino)-3α-(3'-isopropylidene)-lactam-5α-pregn-4β,16β-diol (1), (+)-(20S)-20-(dimethylamino)-3α-(3'-isopropylidene)-lactam-5α-pregn-4β-ol (2),3α-methylsenecioylamino-5α-pregn-20-one (3),3α-methylbenzoylamino-5α-pregn-20-one (4) and (6R, 7E,9R)-9-dimethylamino-4,7-magastigmedien-3-one (5); along with 17 known compounds were identified. Compound 8-10 and 12-22 were isolated from Pachysandra terminalis for the first time.In the anti-tumor metastasis screening experiment of the extracts and compounds, the total alkaloids from the EtOAc extract played a role in anti-tumor metastasis with MDA-MB-231 cell lines. At the concentration of 0.2μg/ml alkaloid compounds 1-9 revealed significant anti-tumor metastasis activity, while they didn't reveal cytotoxic acticity at that concentration.In a screen for antioxidants from the extract of Pachysandra terminalis, the EtOAc extract revealed significant antioxidant activities, and compounds 14 and 22 revealed DPPH free radical scavenging rates with 97.8% and 92%, respectively.
