The study of the chemical constituents of the Plumula Nelumbinis began on the middle of the last century.So far,a variety of alkaloids,flavonoids,polysaccharides,volatile oils et al.have been isolated and purified from Plumula Nelumbinis.The study of its pharmacological activities also have a long history in China.In recent years,lots of pharmacological activities,like reduce blood pressure,anti-arrhythmia,hypoglycemia,antioxidation,immunosuppression,bacteriostasis and anti-cancer,have been found from the alkaloids and flavonoids in Plumula Nelumbinis.However,the research mechanism of chemical constituents and pharmacological activities of Plumula Nelumbinis is still incomplete according to the related literatures,which further exploration need to do.Objective: The aim of this work is to study on the chemical constituents of total flavonoids and alkaloids from Plumula Nelumbinis,combined with antioxidant and anti-inflammatory activities in vivo and in vitro methods,further reveals the pharmacological activity basis of the effective part of Plumula Nelumbinis,and provides reference for its subsequent development and utilization.Method: Firstly,Plumula Nelumbinis was extracted by reflux extraction technology,and the total flavonoids and alkaloids were enriched by cation exchange resin and macroporous adsorption resin.Secondly,HPLC,UV,UPLC-ESI-Q-TOF-MS were used to identified the total flavonoids and alkaloids of Plumula Nelumbinis,and the qualitative and quantitative analysis of total flavonoids and alkaloids were measured.Finally,DPPH,ORAC and FRAP methods were used to analyze the antioxidant activity of total flavonoids and alkaloids in Plumula Nelumbinis;TPA-induced ear swelling model was used to explore the anti-inflammatory activity of total flavonoids and alkaloids in Plumula Nelumbinis.Result: The fingerprints and contents of total flavonoids and alkaloids in Plumula Nelumbinis from 13 different habitats were comparatively studied by HPLC and UV.It was found that the chemical constituents of total flavonoids and alkaloids in Plumula Nelumbinis from different habitats were similar,but their contents were quite different.Thirt-eight flavonoids(Twenty flavonoid C-glycosides,fifteen flavonoid O-glycosides and three aglycones)and Thirty alkaloids(fifteen benzylisoquinoline alkaloids,Five bisbenzylisoquinoline alkaloids,eight aporphine alkaloids,one proaporphine alkaloid and one isoquinolone alkaloid)were identified by UPLC-ESI-Q-TOF-MS method from total flavonoids and alkaloids of Plumula Nelumbinis from Xiangtan,Hunan Province.Two new compounds(isococlaurine-5'-O-pentoside,coclaurine-5'-O-pentoside)were identified for the first time in this study.Conclusion: The analysis of chemical constituents of total flavonoids and alkaloids in Plumula Nelumbinis showed that there were still a lot of active compounds need to be extracted and separated.The study of antioxidant and anti-inflammatory activities provided a reference basis for the use of total flavonoids and alkaloids in Plumula Nelumbinis as effective parts,and also provided a basis for further exploring the pharmacological activities of active compounds in Plumula Nelumbinis.
