[Objective] Large study found that TNF-αin plasma Of obese patients and type 2 diabetes (T2DM) significantly increased. Therefore, obesity and type 2 diabetes is considered an chronic and low inflammatory state. Over-expression of TNF-αin insulin resistance are considered to play a central role in media. But the molecular mechanism leading to insulin resistance is unclear.Using tumor necrosis factor-α(TNF-α) to treat 3T3-L1 adipocytes, several genes were to be observed, including tumor necrosis factor-α(TNF-α) on TNF-αmRNA, adiponectin (ADPN) mRNA, peroxisome proliferator-activated receptor gamma (PPAR-γ)mRNA, interleukin-6 (IL-6) mRNA and interleukin-1β(IL-1β) mRNA expression in 3T3-L1 adipocytes. The influence of Using tumor necrosis factor-α(TNF-α) to treat 3T3-L1 adipocytes, several genes were to be observed, including tumor necrosis factor-α(TNF-α) on TNF-αmRNA, adiponectin (ADPN) mRNA, peroxisome proliferator-activated receptor gamma (PPAR-γ)mRNA, interleukin--6 (IL-6) mRNA and interleukin-1β(IL-1β) mRNA expression in 3T3-L1 adipocytes. The role of TNF-αin the pathophysiological course of obesity-inflammation-insulin resistangce(IR)-type 2 diabetic mellitus(T2DM) were to be explored.[Methods] (1)3T3-L1 preadipocytes was cultured and induced into mature adipocytes. Observed the expressions of PPAR-γmRNA,ADPN mRNA in the differential course of the adipocytes. (2)Observed the expressions of PPAR-γmRNA, ADPN mRNA, TNF-αmRNA, IL-6 mRNA and IL-1βmRNA in mature adipocytes which were treated with high concentration (20μg/L) of the recombinant TNF-αunder diverse phase.[Results] (1)The levels of PPAR-γmRNA,ADPN mRNA expressions were up regulated gradually within the differential course of 3T3-L1 adipocytes. (2)The results of the genes'expressions in adipocytes treated by TNF-αwere as following:The expressions of PPAR-γmRNA and ADPN mRNA decreased with time-dependent manner. The expression of TNF-αmRNA and IL-6 mRNA increased significantly. The expression of IL-1βmRNA didn't show change in statistics.[Conclusion] (1)With the 3T3-L1 preadipocytes being diferentiated into the mature adipocytes, the level of PPAR-γmRNA and ADPN mRNA expression increased gradually along with the differential course.(2)TNF-αpromoted the expressions of TNF-αmRNA and IL-6 mRNA, and inhibited that of ADPN mRNA and PPAR-γmRNA in a time-dependent manner.(3)The inflammation hypothesis on etiologic mechanism of type 2 DM was studied at molecular level in this work, TNF-αplays an important role in the course of inflammation-IR-T2DM.
