Objective: This topic by observing the abdominal aorta caused by the pressure to narrow the ultra-negative rat heart model of mitochondrial adenine nucleotide translocator (ANT) at different times of the dynamic expression, The objectives of our test are as follows:①To investigate the dynamical expression of ANT informs in rats with the pressure overload.②To investigate the relationship between myocardial apoptosis and the dynamical expression of ANT1 in rats with the pressure overload.③To know the effect of the dynamical expression of ANT1 on myocardial energetic metabolism in rats with the pressure overload.④To know the effect of the dynamical expression of ANT at different times in rats with the pressure overload.⑤To know application of metoprolol, captopril, Xin Fu Kang Oral Liquid can affect the oral expression of ANT.Methods: Random allocation was taken to averagely divide 150 SD rats into five groups:Sham operation(SH),Coarctation of abdominal aorta model group (CAA),Captopril Tablets cure group(CAP), MET Metoprolol Tablets cure group(MET) and Xin Fu Kang Oral Liquid group(XFK). Pressure overload myocardial rat model is maked to Some narrowing of abdominal aorta, to take 4 weeks, 8 weeks, 12 weeks of rat cardiac apex, the relative mRNA content of ANT1 and ANT2 in myocardium is detected by RT-PCR and gel electrophoresis imaging.One-way ANOVA is used in statistically significant difference(p<0.05).Result:①CAA group compared with SH group at 4 weeks, 8 weeks, 12 weeks ANT1mRNA content gradually increased, while the ANT2mRNA decreased gradually, there are significant differences(p<0.05).②MET group, XFK group, CAP group than in the CAA group at 4 weeks,8weeks, 12 weeks ANT1, ANT -2mRNA were significantly improved statistically signifycant (p <0.05).③MET group, CAP group, XFK group in 4 weeks and 8 weeks, they have significant differences (p <0.05), while the 12 weeks no significant change (p> 0.05).Conclusion: Both the relative mRNA content of ANT1 and ANT2mRNA vary regularly, we can make the conclusion:①With the increased cardiac pressure overload, Induced apoptosis ANT1mRNA content significantly increased,while, Involved in energy metabolism ANT2mRNA content decreased, these show that the load increased myocardial apoptosis and energy metabolism exist.②Metoprolol, captopril can significantly improve the ANT and ANT2mRNA content, This shows that by improving the activity of ANT metoprolol, captopril can inhibit cell apoptosis and improve of energy metabolism, which may be to improve the pressure overload one of the mechanisms of myocardial injury;③Yi qi wen yang Promoting blood circulation drugs Xin Fu Kang Oral Liquid can improve the pressure overload myocardial ANT1 and ANT2mRNA content, especially in the early stage is better than metoprolol, captopril, indicating Chinese medicine can also inhibit apoptosis and improve energy metabolism and add more options for us in the clinical applications.
