The Experimental Study On Anti-inflammatory And Analgesic Effect Of GU TONG FILM And Skin Drug Acute Toxicity

ObjectiveGUTONG FILM is developed from a proved prescription which is used by Chinese Traumatology of Hubei Hospital of Chinese Medicine to treat Osteoarthritis.This prescription has been in clinical use for more than 10 years with favorable curative effect.This research, via a serials of experiment,is to observe the anti-inflammation and analgesic effects as well as the acute toxicity reaction of GU TONG FILM so as to provide reliable theoretical basis for the clinical use.MethodsMice and rats which meet the requirement of the experiments were randomly divided into four groups,namely,MATRIX OINTMENT group, TONG LUO QU TONG OINTMENT group,low-dose GU TONG FILM group and high-dose GU TONG FILM group,of which the former two are the control groups,the latter two are the experimental groups.After removing their hair on the back in 24 hours later,the mice and rats,based on group,were given MATRIX,TONG LUO QU TONG OINTMENT,low-dose GU TONG FILM and high-dose GU TONG FILM stick accordingly,in which rats given 1/4 stick and mice 1/8 stick. The medicines were applied once respectively on 1st,3nd,5th and 7th days after experiments began.2 hours after the last application,the anti-inflammatory effects of GU TONG FILM were evaluated with the mice's auricle swelling and the rat's toe swelling test respectively caused by xylene and egg white,and the analgesic effects of GU TONG FILM were detected by the hot plate test and the writhing test.40 white guinea pigs were randomly divided into MATRIX group,high-dose group,medium-dose group and low-dose GU TONG FILM group.24 hours after removing their hairs on the back,their skin were abraded by a fine sand paper and treated immediately with one stick of MATRIX,low-dose GU TONG FILM,medium-dose GU TONG FILM and high-dose GU TONG FILM respectively based on groups;and were treated once again 12 hours later.After 24 hours,the residual drugs were washed up. The mucous membranes of skin,hair gloss,limbs activity,breathing,urine,etc. of the guinea pigs were observed for a week;and the death number and the weight of the living guinea pigs were recorded to observe the acute toxicity reaction of the GU TONG FILM on skins.After the experiment,SPSS16.0 was used to deal with the experimental data. Paired data was compared by the t test and multiple sets of measurement data was compared by the q test.ResultsThe test results of xylene-induced the mice's auricle swelling show that:high-dose and low-dose GU TONG FILM and TONG LUO QU TONG OINTMENT can significantly inhibit xylene-induced mouse ear swelling(P<0.01);low-dose GU TONG FILM may affect equally to the role of TONG LUO QU TONG OINTMENT(P>0.01).while the high-dose GU TONG FILM may be better than TONG LUO QU TONG OINTMENT (P<0.01).Test results of egg white-induced the rat's toe swelling show that:GU TONG FILM and TONG LUO QU TONG OINTMENT can significantly inhibit the egg white-induced the rat's toe swelling(P<0.01),but low-dose GU TONG FILM may affect less than 4 hours(P> 0.01);high-dose GU TONG FILM may affect equally to the role of TONG LUO QU TONG OINTMENT(P>0.01).while the effect of low-dose GUTONG FILM may be not as good as TONG LUO QU TONG OINTMENT (P<0.01).The results of the hot-plate test show that:there is no analgesic effect of MATRIX OINTMENT(P>0.01),while the low-dose GU TONG FILM,high-dose GU TONG FILM and TONG LUO QU TONG OINTMENT can significantly increase the pain threshold in mice.(P<0.01).The results of the writhing test show that:the high-dose and the low-dose GU TONG FILM and TONG LUO QU TONG OINTMENT all can reduce the times of writhing of rats which induced by Glacial acetic acid (P<0.01).There is no significant difference between them (P> 0.01).The tests of medication acute toxicity on skins show that:no guinea pigs died,and the surviving guinea pigs are in good condition;the weight change among the groups showed no significant difference (P> 0.01).ConclusionThe topical application of GU TONG FILM can play a significant anti-inflammatory and analgesic effect and no significant acute toxicity action in current dosage,so GU TONG FILM is save and effective in patients with osteoarthritis.
