The Expression And Significance Of T Lymphocyte Subsets And Perforin Of BCG-immunized Mice

Objective:To investigate the expression and significance of perforin and T Lymphocyte Subsets of Mice vaccined with Bccillus Calmette Guerin(BCG).Methods:1.Test animals :Thirty KM mice were divided into two groups randomly(each 15):BCG group and BCG-TB group.2.Take blood:At the beginning of the test,two groups all were injected with 0.3ml Bccillus Calmette Guerin(BCG) via subcutaneous respectively;Every mice in the BCG-TB group was given 0.1ml mycobacterium tuberculosis bacterium suspension(the germ count 109) intravenously after 3 months. For comparison,mice from control group(BCG group)were injected simultaneously with the same volume normal saline via tail vein respectively .Each mice in two groups was taken 1.5-2ml blood via angular vein at the end of 4 month and analyzed T lymphocyte subsets and Perforin by Flow Cytometer respectively. 3. Detection: Surface markers of T lymphocytes were stained with CD3PerCP,CD4FTTC,CD8APC, and intracellular perforin molecules with PE.Analyzing at the total leukomonocyte count,the counts and the percentages of CD3+,CD4+,CD8+,CD4+ CD8+ T lymphocytes and Perforin expressed in T lymphocyte subsets by Flow Cytometer.4. The detection of pathology and bacteriology about lung tissue .Results:1. BCG-immunized mice had not got tuberculosis after the attack of volume and toxicity Mycobacterium tuberculosis .2. The counts of CD3+,CD8+,CD8+%,DP T lymphocytes in BCG-TB group were more than that in BCG group stastically significant (P<0.05).The proportion of CD4+ /CD8+ was less than that in BCG group statistically significant (P<0.05).3. T Lymphocyte Subsets of two groups all showed the increasing of PFN at different degree.4. The counts of CD3+,CD8+,DP T lymphocytes which expressed Perforin in BCG-TB group were more than that in BCG group stastically significant (P<0.05).The proportion of CD4+ /CD8+ expressed Perforin was less than that in BCG group statistically significant (P<0.05).5. The liver,spleen and lung's tissues slices of mice didn't see the tuberculose focus and Mycobacterium tuberculosis by acid-fast staining in BCG-TB group.Conclusions:(1)BCG vaccination can protect the host from tuberculosis when they were infected by Mtb and the performance of immunology is that the counts of CD8+,DP T lymphocyt and the epression of Perforin in T Lymphocyte Subsets increased.(2)CD8+,DP T lymphocyts are the protective immune effect cells against tuberculosis .(3)Increased expression of Perforin in T Lymphocyte Subsets play an important role in anti-tuberculosis.
