Objective:SUMO-1 gene controls the function of proteins which associated with the genesis and development of cancer. Recent studies showed that the expression of SUMO-1 in SMMC-7721 and hepatocellular catcinomas (HCC) tissues expressed marked contract enhancement, The further suggest that SUMO-1 gene is associated with the genesis of cancer. The aims of this study was to evaluate the efficiency of silencing SUMO-1 induced by siRNA in hepatocellular carcinoma cell line SMMC-7721 and to examine if there were any inhibition of Bcl-2 and c-myc expression after being silenced SUMO-1 gene in vitro.Methods:Three strip artificial SUMO-1 siRNA was transfected into SMMC-7721 by means of Lipofectamine? 2000, the final concentration was 100nM. after 24,48,72 hours , extract the mRNA and protein, analysis the expressions of SUMO-1 by RT-PCR and western blot, sereening efficiently siRNA. The SUMO-1 siRNA was transfected into SMMC-7721 by means of Lipofectamine? 2000, after 48hours, analysis the expressions of SUMO-1,Bcl-2 and c-myc by RT-PCR and western blot.Results : Our data showed that the siRNA which sequence is 5ï¹-CAAGAAACTCAAAGAATCA-3ï¹ could significantly silence the expression of SUMO-1 in SMMC-7721.The silencing rate was 9.80%,73.43% and 46.56% at 24, 48 and 72 hours respectively. After being transfected SUMO-1 siRNA, the expressions of Bcl-2 and c-myc were down-regulated significantly. Conclusions: SiRNA is a good manner to silence the expression of SUMO-1 in SMMC-7721 in vitro. Owing to the inhibition of Bcl-2 and c-myc expression induced by SUMO-1 siRNA, SUMO-1 controls the expression of Bcl-2 and c-myc in HCC cell line SMMC-7721.
