Objective: Evaluate the prevalence of HPV16 infection in esophageal squamous cell carcinoma of the Kazakh,Uyghur,Han ethnic group in Xinjiang. To investigate the correlation between the inherited affectivity of the HLA-DR antigen and esophageal squamous cell carcinoma of the Kazakh,Uyghur,Han ethnic group in Xinjiang. To analyse the relationship between HPV16 infectious and HLA-DR antigen in process of esophageal squamous cell carcinoma in Xinjiang. Methods: Polymerase chain reaction(PCR)was employed to screen HPV16 infection statuses of all samples based on type-specific primer for HPV-16 in 355 cases with esophageal squamous cell carcinoma and 208 cases cancer-free controls in the same area. To detect the expression of the HLA-DR antigen in 197 cases with esophageal squamous cell carcinoma of the Kazakh,Uyghur,Han ethnic group in Xinjiang and 197 cases cancer-free controls in the same area by applying to the method of Two_steps of immunohistochemistry. Theχ2 test and the rank sum test were adopted to check the difference in the prevalence of HPV16 infection, expression of HLA-DR antigene between cases and controls by using the statistics software of SPSS 16.0. Results: The total infectious rate of HPV16 was 27.0%(96/355) in 355 cases of xinjiang ESCC, and control group was 19.2%(40/208). There was significant statistical significance between ESCC and control group. The infectious rates of HPV16 of Kazakh,Uyghur,Han ethnic group respectively were 24.4%(44/180),31.7 %(26/82), 28%(26/93),control group was 19.2 %(40/208). There was no significant statistical significance among three groups(P > 0. 05),and there was significant statistical significance between Uyghur and control group(P < 0. 05),but there was no significant statistical significance between Kazakh and Han ethnic group(P > 0. 05). There was no significant statistical significance in age, sex, pathological differentitation and clinical stage of ESCC in xinjiang(P > 0.05). The expressed rate of HLA-DR antigen was 41.1% in 197 cases of xinjiang ESCC, and control group was 7.1%. There was significantstatistical significance between ESCC and control group(P > 0. 05) .In Kazakh,Uyghur and Han ethnic group, there was significant statistical significance compare to control group(P <0. 05),but there was no significant statistical significance among Kazakh,Uyghur and Han ethnic group(P > 0. 05) . The expressed rate of HLA-DR antigen had significant statistical significance between esophagus intraepithelial carcinoma and the invasion esophagus carcinoma,and it had significant statistical significance in TNM clinical stage(P < 0. 05),but it had no significant statistical significance in age, sex and pathological differentitation of ESCC in xinjiang(P > 0.05). The expressed rate of HLA-DR antigen had significant statistical significance with HPV16 positive cases and HPV16 negative cases in 197 cases of xinjiang ESCC,and it had correlation to the infection of HPV16(r =0.639. Conclusion: The infection of HPV16 has a possible relation to esophageal squamous cell carcinoma in Xinjiang. Compare to control group, Uyghur ethnic group has significant statistical significance, Kazakh and Han ethnic group have no significant statistical significance.It may be not enough sample size. The HLA-DR antigen may be one of genetic susceptibility factors of heredity of esophageal carcinoma in xinjiang. The HPV16 infection may be associated with HLA-DR antigen in the development of esophageal cancer in Xinjiang. HPV16 infection associated with TNM clinical stage ,but the HLA-DR antigen are not.They have no diffence in ethnic and are't all associated with age, sex and pathological differentitation.It needs more samples to confirm.
