Objective:To explore the role of HGF,VEGF-C,VEGFR-3and LMVD in theoccurrence and development of human esophageal squamous cell carcinomaby detecting the expression of the factors in normal mucosa and esophagealsquamous cell carcinoma, as well as the relationships of the factors with theclinical pathological characteristics of esophageal squamous cell carcinomapatients such as gender, age,length of tumour, differentiation,depth ofinvasion,lymph node metastasis and TNM stage so as to evaluate therelationships among these factors and prongosis.Methods:50cases of esophageal squamous cell carcinoma (ESCC) and20cases ofesophageal normal mucosa(NM) which were removed and certified bypathology from July,2009to April,2010of the affiliated hospital of ChengdeMedical College were collected. RT-PCR was used to detect mRNAexpression of HGF and VEGF-C,immunohistochemical staining was used todetect protein expression of HGF,VEGF-C and VEGFR-3,LMVD.Results:1The expression of HGFmRNA,VEGF-CmRNA,HGF,VEGF-C,VEGFR-3and LMVD in normal mucosa and esophageal squamous cellcarcinorma.1.1The expression of HGFmRNA,VEGF-CmRNA in normal mucosaand esophageal squamous cell carcinorma.1.1.1The expression of HGFmRNA in NM,ESCC.HGF mRNA in ESCC was higher than that in NM (P <0.01).1.1.2The expression of VEGF-CmRNA in NM,ESCC. VEGF-CmRNA in ESCC was higher than that in NM(P <0.01).1.2The expression of HGF,VEGF-C,VEGFR-3and LMVD in NM,ESCC1.2.1The expression of HGF protein in NM,ESCCThe expression of HGF in ESCC was higher than that of in NM (P<0.01,).1.2.2The expression of VEGF-C and VEGFR-3in NM,ESCCThe expression of VEGF-C and VEGFR-3in ESCC were higher thanthose of in NM(P <0.05, P <0.05).1.2.3LMVD in NM,ESCCLMVD in ESCC was higher than that in NM(P <0.05).2The relationship between HGFmRNA,VEGF-CmRNA,protein ofHGF,VEGF-C,VEGFR-3and LMVD and clinical pathological characteristic ofcolorectal carcinoma.2.1The relationships between expression of HGFmRNA andVEGF-CmRNA.2.1.1The relationships between expression of HGFmRNA and clinicalpathological characteristics of ESCC.The expression of HGFmRNA was positively correlated with lymph nodemetastasis and TNM stage. The expression of HGFmRNA of lymph nodemetastasis group was higher than that of without lymph node metastasis group(P <0.01).The expression of HGFmRNA of TNM stage Ⅲ and Ⅳwas higherthan that of TNM stage Ⅰ andⅡ(P <0.01).2.1.2The relationship between VEGF-C mRNA and clinical pathologicalcharacteristic of ESCC.The expression of VEGF-C mRNA was positively correlated with lymphnode metastasis and TNM stage. The expression of HGF mRNA of lymphnode metastasis group was significantly higher than that of without lymphnode metastasis group (P <0.01).The expression of VEGF-C mRNA of TNMstage Ⅲ and Ⅳw as significantly higherthan that of TNM stageⅠ a ndⅡ(P <0.01).Correlations were not found between HGFmRNA,VEGF-C mRNA and every other characteristics of ESCC such as gender, age,length of tumour,differentiation and depth of invasion.2.2The relationships between expression of protein of HGF,VEGF-C,VEGFR-3and LMVD and clinical pathological characteristics of ESCC.2.2.1The relationships between expression of HGF protein and clinicalpathological characteristics of ESCC.The expression of HGF protein was positively correlated with lymphnode metastasis and TNM stage.The positive expression of HGF protein oflymph node metastasis group was significantly higher than that of withoutlymph node metastasis group (P <0.01).The positive expression of HGFprotein of TNM stage Ⅲ and Ⅳ was significantly higher thanthat of TNMstageⅠ andⅡ (P <0.01).2.2.2The relationship between VEGF-C protein and pathologicalcharacteristic of ESCC.The expression of VEGF-C protein was positively correlated with lymphnode metastasis,TNM stage (P <0.05, P <0.05,P <0.05). Correlations were not foundbetween VEGF-C ptotein and every other characteristics of ESCC such asgender,age,length of tumour,differentiation degree (P>0.05,P>0.05,P>0.05P>0.05,P>0.05).2.2.3The relationships between expression of VEGFR-3protein andclinical pathological characteristics of ESCC.The expression of VEGFR-3protein was positively correlated withlymph node metastasis and TNM stage.The positive expression of VEGFR-3protein of lymph node metastasis group was significantly higher than that ofwithout lymph node metastasis group (P <0.01).The positive expression ofHGF protein of TNM stage Ⅲ and Ⅳ was significantly higher thanthat ofTNM stage Ⅰ andⅡ (P <0.01).There were no relationships between HGF,VEGF-C,VEGFR-3proteinand every other characteristics of ESCC such as gender,age,length oftumour,differentiation and depth of invasion.2.2.4The relationship between LMVD and pathological characteristic of ESCCLMVD was significantly correlated with depth of invasion,lymph nodemetastasis and TNM stage (P <0.01).There were signifycant differencesbetween low-mucous differentiation group and moderate-high differentiationgroup (P <0.05). Correlations were not found between LMVD with patient’sgender,age and length of toumour(P>0.05).3The relationship among HGF,VEGF-C,VEGFR-3,LMVD.3.1The relationship between HGF mRNA and VEGF-C mRNA.The positive relationship could be seen between HGFmRnA andVEGF-CmRNA,(r=0.740P <0.01).3.2The relationship between HGF,VEGF-C and VEGFR-3,LMVD.HGF was significantly correlated with VEGF-C,VEGFR-3and LMVD(r=0.585,r=0.412,r=0.375P <0.01).VEGF-C and VEGFR-3weresignificantly correlated with LMVD(r=0.368,r=0.415P <0.01) and thestrongly correlation could be seen between VEGF-C and VEGFR-3(r=0.514,P<0.01).Conclusions:1The high expression of HGF, VEGF-C and VEGFR-3may play animportant role in the occurrence and development of esophageal squamouscell carcinoma.2HGF,VEGF-C,VEGFR-3was significantly correlated with lymph nodemetastasis and TNM stage in esophageal squamous cell carcinoma. So theycould be a predictive and prognostic factor of ESCC.3There were significantly positive correlation among HGF,VEGF-C,VEGFR-3and LMVD in ESCC. Suggesting that HGF,VEGF-C,VEGFR andLMVD were cooperated in development of esophageal squamous cellcarcinoma and metastasis of lympho tube.
