Objective: To establish Echinococcus multilocularis wister rat model; to search for method of 131I labeled anti-alveolar echinococcosis polyclonal with chloramine T; and to study radioimmunoimaging of 131I labelled anti-alveolar echinococcosis polyclonal antibody in the rat model with Echinococcus multilocularis in vivo. Methods: 60 female wister rats were injected Echinococcus multilocularis through euthyphoria liver biopsy, after 15 weeks vaccination rate were calculated. 24 alveolar echinococcosis rats were randomly divided into A and C groups. B group included 12 normal rats. A and B group rats were injected intraperitoneally 1mCi 131I labelled anti-alveolar echinococcosis polyclonal antibodies, C group rats were injected intraperitoneally 1mCi 131I. All rats were imaged 24, 48, 72, 96 hours later with ECT. And we measured unit weight radioactive counts per minute of three group rats organs after 96 hours.Results: Infection rate of Echinococcus multilocularis wister rat were 62%. The labeled requirement of 131I labeled anti-alveolar echinococcosis polyclonal with chloramine T were 150ug/100ul,131I 3mci/30ul,chloramine T 40ug /40ul,Na2S2O5 80ug /40ul. In A group rats inoculated right lobe of liver alveolar echinococcosis, radioactive parts of the distribution can be seen. And 72 hours alveolar echinococcosis ROI were the highest. After dissection the location were in line. Normal control group, no significant radioactive liver distribution. After 96 hours the execution of three groups rats, A group echinococcus multilocularis in vitro determination of radioactive counts show that the rest of organize T/NT ratios were greater than 1.28 apart from the thyroid and gastric tissue. Conclusion: 131I labelled anti-alveolar echinococcosis polyclonal antibody can be successfully combination with alveolar echinococcosis, for the next step of the way to the use of monoclonal antibodies radioimmunoimaging provides the experimental basis.
