Purpose:Study of classical Chinese medicine Ge Xia Zhu Yu Tang combines with chemotherapy drug Cyclophosphamide used in Hca-f liver cancer mouse damps the lump function and the function mechanism.Material and method:Check 40 health Kunming mice,female and male half .Under sterile condition, collected Hca-f mouse ascites tumor source for the mice,diluted with NS for 2x107 tumor cells/ml,inoculated 0.2ml per mouse to the right side of its armpit under the skin.After 24 hours were randomly divided in model group,CYgroup,GXZY group,CY+GXZYgroup.Treatment of 8 days,the mice were killed ,stripped and weighted tumor weight, calculate the rate tumor.Observe the change of tumor cells in HE staining;see apoptosis by electron microscope ,detected Bcl-2 protein expression by immunohistochemistry .Results:1.The survival status of mice in each group: Model mice skin dryness, not smooth, slow, unresponsive, lack of appetite. CY mice fur bleak, sparse, there is loss, dull, less active, slow response, do not like eating; GXZY shiny fur of mice, mobile and flexible, responsive, more normal diet flexible move,also the mental state the best;CY + GXZY mice fur supple, smooth, lively, responsive, normal diet.2.After drug treatment,Hca-f liver tumou growth in mice are subject to different degrees of inhibition,the inhibition rates were: CY group 44.69%, GXZY group 41.84%, CY+GXZY group 47.75%.3.Through the light microscope,model group can be seen in tumor tissue with tumor group disorder,loss of nomal structure,tumor cell size,shape ,irregular, deeply stained nuclei, has pathological mitotic more common. CY group and GXZY group can be seen in tumor tissue well regulated and pathological mitotic is less than model group. CY+GXZY group arranged in tunor tissue compared with rules,tumor cell size,shape and more consistent,pathological mitoses is much less than the model group.4.Electron microscope, model group, the nucleus was round, large nuclei, nuclear chromatin into high density agglomerate in larger quantities, no apoptotic bodies. CY group, GXZY group, CY + GXZY group of apoptotic bodies could be seen, including CY + GXZY group the largest number of apoptotic bodies.than other group.5.Detected by immunohistochemical staining expression of Bcl-2 protein.The average gray value of model group is 91.052, CY group is 85.846.GXZY group is 87.49, CY + GXZY is 85.31 which is the lowest of all the group.Conclusion:1. Ge Xia Zhu Yu Tang combined with cyclophoamide has increased Hca-f quality of life in the role of liver cancer in mice.2. Ge Xia Zhu Yu Tang combined with Cyclophosphamide can inhibit the Hca-f the role of liver tumor growth in mice.3. Ge Xia Zhu Yu Tang combined with Cyclophosphamide can induce tumor cell apoptosis.4. Ge Xia Zhu Yu Tang combined with Cyclophosphamide can reduced expression of bcl-2 protein.5. Ge Xia Zhu Yu Tang combined with Cyclophosphamide can increase quality of life in Hca-f liver cancer in mice,inhibit tumor growth by inducing tulor cell apoptosis,reducing expression of Bcl-2 protein.
