Objective: To observe the influence of salidroside on rat's liver injury in imitated high altitude condition by hepatic artery ligation(HAL) and possible mechanisms.Method: Rats were classified as ligation group(A),salidroside+ligation group(B),salidroside+ligation+ly294002 group(C) and normal control group.A,B,C group were divided separately into five time as 1h,2h,4h,8h,12h .liver function targets in serum were detected by automatic biochemistry analyser;the content of MDA was detected with TBA(Thibabituric Acid) method;the vigor of SOD was detected with XO(xanthine oxidase) method;pathological change in liver tissue was observed by HE staining;the level of HIF-1αprotein was detected by immunohistochemistry(IHC) and the expression of HIF-1αmRNA was detected with in situ hybridization(ISH);apoptosis in hepatocytes was detected by tunel(TdT-mediated dUTP nick end labeling) method.Results: Hepatic artery ligation(HAL) could cause acute liver injury owing to oxidative stress and apoptosis of hepatocytes.interfere with salidroside can improve rat's liver function targets;lower the content of MDA and highten the vigor of SOD ; abate liver tissue's histopathological change;increase HIF-1α's level both in protein and mRNA;lessen cell apoptosis in liver tissue。conclusions: Salidroside has protective effect on acute liver injury rat caused with hepatic artery ligation(HAL) by opposing oxidative stress and activating PI3K path,up-regulating HIF-1αlevel,inhibiting hepatocyte apoptosis。...
