Objective: To study the effects of cordyceps japonicus capsule prepared by Qinghai Hospital of Traditional Chinese Medicine on oxidative stress molecules,inflammatory factors and cardiac function indexes of Wistar rat model under high altitude hypoxia,and to explore the protective effect and mechanism of cordyceps japonicus capsule on myocardial tissue injury in rats.Methods: Sixty Wistar rats were purchased and fed in cages for 3 days in Xining,Qinghai province(2,200 m above sea level).They were randomly divided into blank control group,model group,salidroside group,cordyceps japonicus capsule low-dose group,sordyceps japonicus capsule medium-dose group and cordyceps japonicus capsule high-dose group,with 10 rats in each group.To medicine group dosage per kilogram of body weight and bulk drug lavage,blank group and model group was given distilled water to fill the stomach volume,continuous dosing in 30 days,in addition to the blank group,the rest of the group of rats to the point of of qinghai province He Na County(at an altitude of 3700 m),in high altitude low oxygen alpine plateau low pressure hypoxia model is set up under the natural environment,After feeding for 48 hours,the animals were anesthetized and killed,and blood and heart tissue were collected.HE staining was used to observe the changes of cardiac histomorphology and compare the degree of cardiac injury in each group.Serum and plasma were separated by centrifugation,and the contents of MDA,TNF-α,IL-6,CK and LDH in serum were determined by ELISA.The contents of CK and LDH in supernatant were determined by ELISA after homogenization.Total SOD activity detection kit(WST method)was used to detect SOD activity.Results:1.Compared with blank group,myocardial cells in model group were disordered and morphologically changed,with elongated nucleus,serious cell damage,severe fibrosis of interstitial cells and infiltration of a large number of inflammatory cells.Compared with the model group,the morphological changes of myocardial cells in the treatment group were lighter,and the inflammation degree of myocardial cells in the high,medium and low dose groups of cordyceps capsule was successively increased.The degree of myocardial injury was similar between the high-dose cordyceps capsule group and salidroside group.2.Compared with blank group,SOD content in serum of model group was decreased,MDA,IL-6,TNF-α,CK,LDH contents in serum and CK,LDH contents in heart homogenate were increased(P<0.05).Compared with model group,SOD content in serum of cordyceps capsule group was increased,MDA,IL-6,TNF-α,CK,LDH contents in serum and CK,LDH contents in heart homogenate were decreased(P<0.05).3.The levels of SOD in cordyceps japonicum capsule group were high dose group > medium dose group > low dose group,and the levels of MDA,TNF-α,serum CK,LDH,heart homogenate CK and LDH were high dose group < medium dose group < low dose group,with statistical significance(P<0.05).The levels of serum IL-6 in cordyceps japonicus capsule groups were high dose group < medium dose group < low dose group,and there was no statistical difference between medium dose group and high dose group(P>0.05).There was statistical difference between low dose and high dose(P<0.05).Compared with blank group,there was no significant difference in serum CK level between cordyceps japonicus capsule high-dose group and salidroside group(P>0.05).There were no significant differences in the levels of SOD,MDA,IL-6,TNF-α,CK,LDH in serum and CK,LDH in heart homogenate between cordyceps japonicus capsule high-dose group and Rhodiola group(P>0.05).Conclusion:1.The high-altitude hypoxia rat model can be successfully established in Henan County,Huangnan Prefecture,Qinghai Province at an altitude of 3700 m.2.American Ginseng cordyceps capsule can prevent hypoxia reaction caused by altitude hypoxia by improving free radical metabolism,improve cardiac function by alleviating inflammation,and alleviate myocardial cell damage caused by altitude hypoxia.
