Artemisia annua is Asterales Asteraceae Artemisia annua Artemisia species of subspecies of Big Head for the annual dicots. It was reported that, Artemisia annua with antimalarial, antibacterial, anti-parasitic, antipyretic, blood pressure and other biological activities, Artemisia polysaccharide is one of its main active ingredient, but few research on the pharmacological activity of Artemisia polysaccharides is reported. This research study on anti-tumor effects of polysaccharides from Artemisia annua by the way of S180 tumor in mice models , in order to provide a reference for the further determine on the mechanism of its anti-tumor.The RSA software is used for extraction technology of total polysaccharides in Artemisia annua by ultrasonic wave. The results showed that, the perfect conditions of ultrasonic extraction for Artemisia annua polysaccharides are as follow: extraction temperature is 52.4℃, extraction time is 36.0min, ultrasonic power is 87.0W and ratio of liquid to solid is 27.8ml.g-1. Parallel experiment is replicated three times, three experiments Artemisia polysaccharides are 16.19%, 16.28% and 16.40%, the average yield is 16.29%, the predictive best polysaccharides extraction rate is 14.23 %,and the relative error is 0.21 %. This shows that the response surface model can simulate and predict the extraction conditions of Artemisia annua.Secondly, S180 tumor mouse model is used to test Antitumor Activities of the extraction from Artemisia annua in vitro, through the plasma and liver tissue of mice in the SOD activity, GSH-Px activity and MDA content and other parameters measured, the results show that Artemisia polysaccharides have no significant cytotoxicity, which can effectively induce lymphocyte proliferation and transformation; In addition, Artemisia polysaccharide can effectively contribute to lymphocyte secretion of IL-2, IFN-γ, promote the secretion of macrophage TNF-α, and with the dose gradually increasing role in promoting increased work results and lentinan considerable; can say that the anti-tumor activity of Artemisia polysaccharides are likely related to its immunomodulatory activity, therefore, in-depth study of immunomodulatory activity of polysaccharides from Artemisia annua will help study the antitumor mechanism of polysaccharide of Artemisia annua, which is a new drug provide a theoretical basis for research and development. Then, through the establishment of S180 tumor model in vivo, measured Artemisia polysaccharide anti-tumor activity in vivo results showed that: in a certain concentration range, obtained by the response surface analysis of the ultrasonic extraction of Artemisia polysaccharides show some anti-tumor effect; and with the increasing concentration of Artemisia polysaccharides in vivo tumor inhibition rate increased gradually, showing a dose - response relationship; and when the concentration of Artemisia polysaccharides increased to 150 mg / kg.d, the inhibitory rate of 56.7%, and cyclophosphamide inhibited tumor growth rate was close, indicating Artemisia polysaccharide could significantly inhibit the growth of mouse S180 tumor. In addition, the results also show that Artemisia polysaccharides can stimulate the immune function and lower organ weights returned to normal levels; the same time, Artemisia polysaccharide also shown in mice serum and liver tissue SOD activity and the GSG-Px activity to promote the role of , and the formation of malondialdehyde less, which also shows Artemisia polysaccharides can increase the tumor-bearing mice the activity of antioxidant enzymes in blood, reduce lipid peroxide content.Finally, acute toxicity, subacute toxicity and chronic toxicity of Artemisia polysaccharides toxicological studies carried out to ensure that Artemisia polysaccharides as drug treatment of cancer using safe and nontoxic, so as to further development of Artemisia polysaccharides provides some reference value.
