Objectiv: The aim of this study was to optimization the extraction and purification process of Lycopus lucidus polysaccharide, discussed the pharmacological activity of the antioxidant.Methods:(1) The content of polysaccharide in the extract was measured by sulfuric acid-phenol method,it was used glucose as standard substance.(2) The extraction rate of polysaccharide from lycopi herba was used as index, through investigated the influencing factors of water extraction, included the temperature, time, and solid-liquid ratio, then determined the optimal extracting technology by the response surface model establishment of Box-Behnken design.(3) The extraction rate of polysaccharide from lycopi herba was used as index, through investigated the influencing factors of supersonic extraction, included the temperature, time, solid-liquid ratio and ultrasonic power, then determined the optimal extracting technology by orthogonal experiment.(4) The extraction rate of polysaccharide from lycopi herba was used as index, through investigated the influencing factors of enzyme extraction, included the enzyme species, p H value, temperature, enzyme dosage, time and solid-liquid ratio, then determined the optimal extracting technology by the response surface model establishment of Box-Behnken design.(5) The weight score from polysaccharides retention rate, decoloration rate, protein prolapse rate was used index, through investigated the influencing factors of the purification results, included the different types of macroporous resin, injecting concentration, the dosage of washing, then determined the purification process technology by the response surface model establishment of Box-Behnken design.(6) The liver brain tissue T-SOD activity, GSH-PX activity, CAT activity, the content of MDA as indexes, Vitamin C effervescent tablets as a positive control medicine, then investigate gavage three different dosages of zeeland polysaccharide effect on aging model mice by intraperitoneal injection caused galactose, these three doses were 100mg/(kg·d), 200mg/(kg·d), 300mg/(kg·d).Results:(1) Set up the standard curve of sulfuric acid-phenol method was as follows: Y=76.591X-0.0846, r= 0.9997.(2) The optimum water extracting technology as follows : the extracting temperature was 70℃, the extracting times was 2, each times lasted 2 hours, solid-liquid ratio was 30, the extraction rate of polysaccharide can reach 2.35%.(3) The optimum supersonic extracting technology as follows: ultrasonic power was 560 W, temperature 60 C, time 20 min, solid-liquid ratio was 30, the extraction rate of polysaccharide can reach 2.31%.(4) The optimum enzyme extracting technology as follows: the mixed enzyme dosage was 2.3%, time was 1h, solid-liquid ratio was 30, the p H value was 4, the extraction rate of polysaccharide can reach 2.033%.(5) The optimum purification technology as follows: the sample concentration was 0.03g/m L, the flow rate was 1.1m L/min, the elution volume is 40 m L, polysaccharide retention rate can reach 69.21%, the decolorization rate can reach 60.24%, proteinremoval rate can reach 75.67%.(6) The polysaccharide of lycopi herba has the pharmacological activity of antioxidation in mice body.Conclusion: It’s a stable technology obtained from this experiment, have a higher progressiveness; lycopi herba polysaccharide antioxidant activity is worthy of further development.
